因B型肝炎病毒(HBV)感染而引起的病毒性肝炎在台灣是一個嚴重的公共衛生問題。以此病毒的表面抗原(HBsAg)製成疫苗,以核心抗原(HBcAg)製成檢定試劑,皆為解決此病毒為害人類的醫療策略。
本實驗乃是嘗試利用基因表現載體(expression vector): pLG669Z, pHC624-iqI-tac,及基因重組(recombinant DNA)技術,分別在酵母系統及大腸桿菌系統中,選殖表面抗原基因及核心抗原基因,並測其表現,以為生產此二抗原之先導實驗。

dc.description.abstractSummary
Hepatitis B virus (HBV) infection is a serious public health problem in Taiwan. Hepatitis B surface antigen (HBsAg) has been successfully used as vaccine for preventing hepatitis B. Both HBsAg and HBcAg are useful reagents for the screening and diagnosis of HBV infection.
The current state of art of obtaining sufficient amounts of hepatitis B surface antigen and hepatitis B core antigen is cloning and expressing the corresponding genes in suitable vectors.
In this research, HBsAg gene-containing DNA fragment prepared by various restriction endonuclease digestion and/or Bal 31 exonuclease treatment has been cloned into E. coli and yeast shuttle vector pLG669Z. HBcAg gene-containing DNA fragment prepared by the similar methods has been cloned into E. coli vector pHC624-iqI-tac.
The expression of both HBsAg and HBcAg is going to be analyzed elsewhere.
