dc.description.abstract | 淋巴素的研究對於吾人瞭解免疫系統的交互作用及調節作用中扮演著重要的角色。 本文以一個新的方向來研究人類淋巴素。PHA刺激的人類淋巴細胞已知可分泌出許多不同的淋巴素於培養液中。因此使用經PHA刺激的人類淋巴細胞的培養液為抗原來產生抗體,並以此培養液來產生35S-methionine標示的淋巴素。令人驚異的是:以兔子的抗淋巴素抗體對剛分離的人類周邊血液淋巴細胞具mitogenic effect,且此效應主要是對應於T淋巴細胞。這個具mitogenic效應的抗體也被用來探測經〔35S〕-methionine標示的細胞表面抗原和淋巴素,使用的為Fluorograph的技術,結果發現在PHA刺激過的細胞表面和培養液中,均測到一個80Kd的分子。 無論由細胞表面抗原或培養液中著手,都極難獲得足夠量的淋巴素來做進一步的研究。因此,必須將淋巴素的基因殖入之λgt-11載體中,且用抗體當探測子來分離出此基因,如此才能產生大量的淋巴素以進一步測定其生化及免疫性質。 | zh_TW |
dc.description.abstract | Lymphokine research plays a key role in the understanding of the interactions in the immune system network. A new approach has been set up for investingating human lymphokines. PHA-stimulated human lymphocytes elaborate a number of distinct lymphokines in the culture media. The serum-free, PHA-conditioned human lymphocyte media has been used for producing rabbit antibodies and for generating (35S)-methionine-labeled lymphokines. Suprisingly the rabbit antibodies have mitogenic effect on freshly isolated human peripheral blood lymphocytes This mitogenic effect is specific for T cells The mitogenic rabbit antibodies have been used for detecting (35S)-methionine-labeled cell surface antigenes as well as lymphokines using fluorographic techniques. An 80Kd polypeptide has been detected only in the PHA-stimulated lymphocyte surface as well as in the conditioned media It is extremely difficult for obtaining sufficient quantities of either cell-surface antigen or lymphokines for studies Therefore, cloning the genes of lymphokines into λgt-11 and using antibodies as probes for detecting these genes are essential for producing large amounts of lymphokines and for studying the biological and immunological properties. | en |
