摘要: | 大豆白化幼苗分別以5-fluorouracil(FU), actinomycioD (AD)及 cycloheximide (CY)處理。AD及CY處理組,對根部細胞延長分別有32%及86%的抑制效果,但FU處理組則無顯著的抑制作用。切取各處理組之大豆白化幼苗根部延長區內的組織,分析蛋白質及核糖核酸含量,發現蛋白質含量和細胞延長有密切的關係。藉電泳膠體分析法,並配合放射性同位素追蹤,比較各種處理組蛋白質類型的變化,發現AD及CY處理組分子量在23,000 daltons的蛋白質帶都消失。以(上標 3)^H-1eucine 做追蹤劑,測定植物體蛋白質新合成的能力,發現三種抑制劑對根部延區組織吸入(uptake) (上標 3)H-leucine,及併入(incorporat-ed)蛋白質的量都有不同的影響。若以CY處理,對(上標 3)^H-leucine之併入蛋白質有顯著抑制作用,以AD處理則有促進作用,且FU處理組對(上標 3)^H-leucine 的吸入組織及併入蛋白質都有促進作用。以(上標 3)^H-uridine做標誌測定核糖核酸新合成能力,發現三種抑制劑的處理都會抑制(上標 3)^H-uridine併入核糖核酸。測量三種處理對β-galactosidase活性之影響,由結果得知該酵素的活性因FU及CY處理而被抑制,但它的活性不被AD抑制。但根部的延長不為FU處理所抑制影被抑制。AD處理有32%被抑制。而CY處理有86由這些結果可推測β-galactsidase並不參與細胞延長過程中,細胞壁鬆弛的作用。 60-hour old etiolated soybean seedlings were treated with 325μg/ml 5-fluorouracil (FU) or 20μg/ml. actinomycine D (AD) or 2μg/ml cycloheximide (CY) for 12 hours and the cell elongation and biochemical changes in the elongation zone of intact root were examined. The inhibition of AD and CY on cell elongation were 32% and 86% respectively. Analyzing the relationship of RNA and protein content in the elongation zone after each treatment and the cell elongation, the results indicated that ther is a close relationship betweem the protein content and cell elongation. The control and FU-, AD-, and CY-treated tissues were labeled with 3^H-leucine for 8 hours, and the newly synthesized proteins from each tissue were analyzed by l2.5% SDS polyacrylamide gel electrophoresis. From the auto-radiograph showed that the protein band at the molecular weight of 23,000 daltons was disappearance after the AD and CY treatment. The comparison the effect of each inhibor on the uptake and the incorporation of 3^H-leucine into protein in the tissue, the results showed that the CY severly inhibited the 3^H-leucine incorporation into protein; FU stimulated these incorporation; and both FU and AD enhanced the uptake of 3^H-leucine into tissues. Using the 3H-uridine incorporation studies, the RNA synthesis was inhibited by all of protein and RNA synthetic inhibitors. The percentage of inhibition of FU, AD, CY were 75.0%, 62.7%, 73.3%, respectively. The β-galactosidase activity on the root tip was severly inhibited by PU, but this inhibitor had no significant inhibition on cell elongation. On the other hand, AD did not inhibit the β-galactosidase activity but this inhibitor did inhibit the cell elongation. From these exrerimental data suggested that β-galatosidase did not play an important role in cell elongation. |
