dc.description.abstract本研究之目的乃在於建立鯉魚及泥鰍之鰭細胞株,並研究此二細胞株之酵素染色及細胞表面顯微構造。
鯉魚及泥鰍鰭細胞在15個月間,各培養了105代和67代,並分別命名為CF及LF。此二細胞株最適宜的生長條件為含20%牛犢血清之Leibovitz'sL-15培養液,溫度在28?31℃之間。細胞表面附著效應值,LF為23?28%,CF為11?22%。LF及CF細胞皆為表皮樣細胞,CF細胞的細胞密度為120,000cells/平方公尺,而LF細胞則為48,000cells/平方公尺。CF及LF細胞株對於魚類病毒EVEX和EVA有感受性,而對其他包括EVE、IPNV、IHNV和VHSV等病毒無感受性。
CF及LF細胞在培養過程中,酸性磷酯?、鹼性磷醋?、腺?三磷醋?、琥珀酸去氫?、葡萄糖一六一磷酯?及細胞色素C氧化?等六種酵素的呈色反應會隨培養時間的增加而加深。其中以酸性磷酯?在CF細胞自體溶蝕時的反應最顯著。
CF及LF細胞在培養過程中,細抱外型會改變:CF細胞會有自發性的自禮溶蝕現象,而培養在37℃時,細胞會重疊且加速退化;LF細胞則由多樣化的外型趨於單一的多角型。這些細胞表面微細構造之連續變化情形,已分別由掃描電子顯微鏡,得到具體的觀察。
