摘要: | 當菸草栽培品種威斯康辛38號(Nicotiana tabacum L cv. Wisconsin 38)之花藥培養於改良式MS培養基時,正值第一次有絲分裂的小孢子,迅速分裂形成一個營養核和一個生殖核,此後營養核能夠不斷地進行細胞分裂,形成多細胞體的構造,而生殖核通常不分裂或只分裂一次便停止了,將來宿存於多細胞體中或退化掉。而水稻栽培品系新竹糯4號(Oryza sativa L. cv. Hsinchu 4)之花藥培養於含有適當荷爾蒙濃度之改良式MS培養基時,其小孢子的發育型式共有三種。其一是中期單核的小孢子經由第一次有絲分裂後,可能產生兩個相等的細胞核,將來這兩個細胞核分別地進行細胞分裂,再共同組成多細胞體。另二種型式是產生一個營養核和一個生殖核,將來可能只有營養核可以繼續分裂下去,或者營養核和生殖核皆具有分裂能力,共同組成多細胞體。本研究擬就這兩種作物花藥培養中,小孢子之發育型式做一些討論和比較。 The pathways of androgenesis in anthers of tobacco (Nicotiana tabacum L. cv. Wisconsin 38) and rice (Oryza sativa L. cv. Hsinchu 4) were studied. When anthers of tobacco containing microspores at the first mitotic stage were cultured on modified MS medium, the first division of the microspores gave rise to a large vegetative and a small generative cell. It was shown that the vegetative cell played the major role in embryogenesis whereas the generative cell and its few derivatives degenerated. If anthers of rice containing mid-uninucleate microspores were cultured on a modified MS medium supplemented with 4 mg/l NAA and 2 mg/l kinetin, the first division of the microspores was either symmetrical or asymmetrical. In the former case of microspores the two similar nuclei divided independently and both contributed to the formation of multicellular units. In microspores which differentiated into a vegetative and a generative cell, continued division of the vegetative cell or of both cells results in the formation of two different types of multicellular units. Discussion and comparison of the segmentation patterns in microspores of tobacco and rice during anther culture were made. |
