dc.description.abstract | 在豬腦下腺作成的丙酮粉末或酸性丙酮粉末,分離出豬的催乳激素,經分子大小篩選及離子交換樹脂的純化。電泳分析得到多條色帶,此現象表示其為非均質,多數催化激素均有此現象。再由分子量,氨基酸分析,N端的蛋白質分析,都與其他作者的報告相符。除了以上的化性實驗外,我們還用鴿子束囊作生物活性鑑定;由牛腦下腺得到催乳激素,在兔子內所產生抗體發現不會與我們萃取之產物相反應。 脫氨的豬腦生長激素也在本實驗中萃取出來,它的化性幾乎與未脫氨的生長激素一致:有相同分子量,N端蛋白質分析,氨基酸分析,在酸性電泳一致而鹼性電泳移動較快,且作有關生物活體及體外的活性實驗;由脫氨生長激素產生的抗體不能與未脫氨的生長激素相反應,此亦在本實驗研究中發現。 | zh_TW |
dc.description.abstract | Porcine prolactin has been isolated from acid-acetone powder and acetone-powder. It is purified by gel-filtration and ion-exchange chromatography. In disc electrophoresis it gives multiple bands which is normal for this hormone. The molecular weight, amino acid composition and N-terminal analysis are in agreement with the reported values. It is active in the pigeon crop sac assay. This porcine hormone cannot react with the antiserum against bovine prolactin in the double diffusion test. Deamidated porcine growth hormone has been obtained which shows all the chemical properties expected for a deamidated growth hormone: same molecular weight, same N-terminal residue, same composition, same mobility in acidic disc electrophoresis but different in basic system. It is able to induce ornithine decarboxylase in vivo and in vitro. An antiserum against it has been raised which can not recognize the native porcine growth hormone. | en |
