| dc.description.abstract | 本報告乃在?述有關吳郭魚卵巢細胞株To-2之建立。此細胞株在添加了10%牛犢血清之Lei-bovitz’s L-15培養液中,於31±1℃的條件下已繼代培養了75次。To-2細胞株的形態為單核及多核之表皮樣細胞。分析100個分裂中期To-2細胞的染色體數目,結果顯示染色體數目的分佈範圍自24至76呈雙向分佈,而以含40個染色體之細胞最多。於低密度細胞培養時,細胞表面附著效應(Plating Effeciency)約12.5%。比較To-2細胞在含2%,5%,10%,20%等四種牛犢血清濃度的培養液中之生長情況,結果顯示其生長速率隨著血清濃度的升高而增高。To-2細胞可生長的溫度範圍自15℃至37℃,而於31℃的條件下生長情況最好。本實驗亦證實了To-2細胞株可接受EVA、EVE、EVEX及IPNV四種病毒,且EVE及IPNV在To-2細胞內之複製情形良好。To-2經鑑定沒有細菌、真菌及黴菌質(Mycoplasma)的污染。 | zh_TW |
