摘要: | 本論文共分兩大部分: 第一部分以靈芝(Ganoderma lucidum)來進行深層液態發酵培養,探討碳源、攪拌速度、通氣有無、高溫高壓滅菌是否對靈芝多醣體(βglucan)的產量造成影響。發現以5%葡萄糖為碳源及0.5%酵母抽出物(yeast extract)為氮源的液態培養液,在第七天之後收成可以獲得較佳及較符合經濟效應的靈芝多醣體。以此培養成分為基本液態培養液,發現在300rpm的轉速並給予通氣塞培養的靈芝多醣,有較高的多醣產量。在高溫高壓滅菌之後,發現靈芝多醣的多醣成分並沒有顯著的降低。 第二部分以不同濃度靈芝多醣體餵與ICR小白鼠,測試其生理活性。在SOD部分,實驗組結果顯示,小白鼠的肝臟及脾臟在一週後就開始有顯著的提升,38天的實驗期間與對照組比較有明顯的上升,其中低劑量組的效果較佳。在脾臟中及血液中淋巴細胞的數量分析部分,以流式細胞儀進行分析,結果顯示,餵與小白鼠靈芝多醣體的實驗組,在淋巴細胞的總量上有顯著的提升,高劑量組和低劑量組在T細胞和B細胞的數量比例上則有所不同。 The present study consists of the following parts: The first part aims to investigate the ability for hyphal stage of Ganoderma lucidum in the production of β-glucan in vitro. The optimum culture medium for the culture of hyphal Ganoderma lucidum was found in a medium containing 5% of glucose, 0.5% of yeast extract. It was also found that aeration, moderate stirring (300rpm) and suitable pH range for the culture system may result an increase of β-glucan production. In the second part of experiment the changes of physiology characters in various tissues of ICR mice were detected when β-glucan was orally ingested with two different dose level during experiment. The results obtained on this experiment revealed that significant enhancement of SOD in liver and spleen was obtained one week with β-glucan derived from Ganoderma lucidum, and the effect was continued for all experiment duration. The SOD effect of low dose level was better than high dose level. The amounts analysis of lymphocytes in blood and spleen was detected with flow cytometer (FACS). The results obtained on this experiment revealed that significant increase of lymphocytes in blood and spleen was obtained when β-glucan derived from Ganoderma lucidum was used. The ratios of T cells to B cells of lymphocytes in blood and spleen respectively were different from high dose level and low dose level. |
