| dc.description.abstract | 摘要 本研究擬分析廣東住西縓蟲(Angiostrongylus cantonensis)之醣解酵素活性,以瞭解其葡萄糖代謝之可能途經。 實驗結果顯示廣東住西縓蟲成蟲體內可分析出下列酵素之顯著活性:葡萄糖?(glucokinase)磷酸葡萄糖異構?(phosphoglucoisomerase),磷酸果糖激?(phosphofructokinase),醛縮?(aldolase),磷酸三碳醣異構?(triosephosphate isomerase),3-磷酸甘油醛去氫?本(glyceraldehydes 3-phosphate dehydrogenase),磷酸甘油激?(phosphoglycerokinase),磷酸甘油酸轉位?(phosphoglyceromutase),烯醇?(enolase)及丙酮酸激?(pyruvate kinase)。而青年期蟲體內也可分析出上列大部分酵素之顯著活性,此結果証明瞭葡萄糖至丙酮酸之分解,可能遵循著Embden-Meyerhof pathway進行。 本研究同時分析各個醣解酵素之催化反應是否因特殊因數之存在而激發其進行,實驗結果顯示:Adenosine-5'-triphosphate可激發葡萄糖激?,3-磷酸甘油醛去氫?及磷甘油酸激?之催化反應的進行。Glucose-6-phosphate及fructose-1-,6-diphosphate則分別可激發6-磷酸葡萄糖去氫?及醛縮?之作用進行,而fructose-6-phosphte則可激發磷酸葡萄糖異構?,磷酸果糖激?之作用的進行,同時磷酸三碳醣異構?則受glyceraldehyde-3-phosphate之激發而可進行催化之作用。丙酮酸激發則受Adenosine-5'-diphosphate激發。 本研究並進一步分析丙酮酸利用過程之酵素活性,分析出4種則具有顯著活性之酵素:乳酸去氫?(lactate dehydrogenase),丙酮酸脫羧?(pyruvatedecarboxylase),乙醇去氫?(alcohol dehydrogenase),丙酮酸去氫?(pyruvate dehydrogenase)。由於這4種酵素的存在說明廣東住血縓蟲之丙酮酸成乙醇代謝有3種可能的途經:1.還原?乳酸,2.發酵成乙醇,3.轉變?乙醯輔?A(acetyl Co A),而進入三羧酸循環(Tricarboxylic acid cycle)。 此外,磷酸甘油可不可能去氫?(glycerophophosphate dehydrogenase),及6-磷酸葡萄糖去氫?(glucose 6-phosphate dehydrogenase)在分析過程中,也出現顯著之活性。 | zh_TW |
