| dc.description.abstract | 在真核細胞中,hnRNP 蛋白質是一群結合到 hnRNA 上的巨大多蛋白複合體,這些 hnRNP蛋白質參與了pre-mRNA 的切割和 mRNA 的運送過程,其中 hnRNPK 與 hnRNPEl/E2是主要結合到poly-C 核酸序列的蛋白質。 急性發炎反應時,C/EBPβ是最主要調控急性反應基因表現的轉錄因數,而 hnRNP K 在 C/EBPβ調控的基因活化上是作為一負調節者,在這篇論文中,我找到另一個poly-C 結合蛋白-mCBP也可以抑制 C/EBPβ誘發的基因活化現象,並且發現登革熱病毒的核心蛋白可以與 hnRNP K 或 mCBP 產生專一性的交互作用,此外,登革熱核心蛋白可以減少 mCBP 對 C/EBPβ誘發的基因活化的抑制效果,但不會影響 hnRNP K抑制C/EBPβ之效果。 | zh_TW |
| dc.description.abstract | In eukaryotic cells, nascent RNA transcripts are associated with Large, multiprotein complex called heterogeneous nuclear ribonucleoprotein complexes (hnRNPs). These hnRNPs participate in the processing of pre-mRNA and in the export of mRNAs from the nucleus. Among these hnRNPs, hnRNP K and hnRNP E1/E2 are known to be the major poly-C binding proteins. Transcription factor C/EBPβhas been known to regulate a wide array of genes including those involved in the acute-phase response. It has been shown that hnRNP K is a negative regulator of C/EBPβ-mediated agp gene activation. In this report, I demonstrated that mCBP is another negative regulator of C/EBPβ-mediated agp gene activation. I also characterized the interaction between Dengue virus core protein and hnRNP K or mCBP. Furthermore, Dengue virus core protein can relieve the suppression effect of mCBP but not hnRNP K on the C/EBPβ-mediated agp gene activation. | en |
