摘要: | 組蛋白修飾會影響染色質的結構及調節基因的表現。阿拉伯芥含JmjC功能域組蛋白去甲基酶JMJ29屬於KDM3/JHDM2類,在過往研究中普遍認為KDM3/JHDM2類蛋白具有組蛋白H3賴氨酸9 (H3K9) 去甲基酶的功能。在本研究中發現KDM3/JHDM2類中的JMJ27、JMJ28和JMJ29彼此之間在細胞核有交互作用,而且JMJ29確實擁有去除組蛋白H3K9甲基化的活性。jmj29突變體在長日照及短日照的條件下都有提早開花的表現型,且其葉片與莖上有較少的毛狀體(trichome),但毛狀體的結構並未受到影響。調控毛狀體發育的轉錄因子GL1、GL3和GL2在jmj29突變體的基因表現相較於野生型有顯著性下調。另外,JMJ29會直接結合在GL3上,GL3上的H3K9甲基化程度在jmj29突變體中有顯著性提升。此外JMJ29蛋白定位於在細胞核中,並且會與GL1有交互作用。這些結果說明JMJ29可能透由調控組蛋白去甲基化的方式來影響GL3基因表現,並參與阿拉伯芥毛狀體生長發育。 Histone modifications play a fundamental role in regulating chromatin structure and gene expression. JmjC domain-containing histone demethylases (JDHM) remove the methyl groups from methylated lysine residues in Arabidopsis. The histone demethylase JMJ29 belongs to the KDM3/JDHM2 group and may have H3K9 demethylation activity. In this study, we found that the members of the KDM3 group proteins, JMJ28 and JMJ29, can interact with JMJ27. JMJ29 has demethylase activity specific to H3K9 methylations. The jmj29 mutant plants display an early flowering phenotype. Furthermore, jmj29 mutant plants show reduced trichome numbers compared to wild-type. The expression levels of the genes encoding the trichome initiation factors GL1, GL3 and GL2 are decreased in jmj29 mutants compared to wild type. JMJ29 can directly target to the GL3 locus and the level of H3K9me2 on GL3 is increased in jmj29 mutants. Furthermore, JMJ29 is localized in the nucleus and can interact with GL1 in the nucleus. Taken together, these results indicate that JMJ29 is involved in trichome development by affecting the H3K9me2 level on the GL3 locus. |
