| dc.description.abstract | 為了瞭解寄生蟲的性質,包括基因型和血清型,以及疫苗可施用的魚種範圍,
本研究收集台灣與大陸閩南地區白點徵狀病魚。首先,從寄生蟲囊孢選殖核醣體 去氧核醣核酸(rDNA)序列,鑑定海水白點蟲,其次利用rDNA 序列分析不同來源 寄生蟲之親緣關係及基因型別。再選殖制動蛋白去氧核醣核酸互補序列(iAg cDNA sequence),分析不同來源寄生蟲免疫原之親緣關係,提供開發多價疫苗的資訊。 最後,分析纖毛帅蟲血清型,作為評估疫苗功效以及追蹤傳染病源之依據。 從2007 年迄2012,從台灣和中國大陸閩南地區取得 78 尾病魚,收集到 47 株寄生蟲,選殖、定序並比對其囊胞體部分 18 S、ITS-1、5.8 S、ITS-2 以及部分 28 S rDNA 序列,鑑定為海水白點蟲。分析被感染的 78 尾病魚,分類上屬於 31 屬42 種,包含食用魚和觀賞魚。分析本研究選殖的 47 條海水白點蟲 rDNA 序列 以及21 條NCBI參考株 rDNA 序列之親緣關係顯示存在五個群簇::群簇 I (大陸地 區分離株)占54 %;群簇 II和群簇 IV(美國和日本分離株)各占16 %和 15%;群簇 III (南海以西地區分離株) 占9 %;群簇V (高雄、澎湖特有分離株)占 3%;而無法 分群的Aus. B與Aus. C 兩株則分別各佔1 %。因此推測海水白點蟲的分化,導致 品系間序列的差異,除地理因素影響外,可能與鹽度有關,而與魚種或溫度無關。 選殖、定序並排列比對 15 條iAg 胺基酸序列發現第 126 個aa 位置上為終結 密碼子;另外,每條 iAg 胺基酸序列中終結密碼子的個數與位置都坐落在不同點, 顯示iAg 胺基酸序列的變異性。雖然 iAg具變異性,本研究鑑定之 092208_Hm_TT iAg 較接近日本的G37 iAg (相似度89.8%) ,而111708_Cs_TT iAg則較近日本的G32 iAg (相似度 87.6%)。親緣關係顯示iAg存在三個群簇。 免疫螢光染纖毛帅蟲發現兔抗血清僅辨認部分纖毛帅蟲,而非全部纖毛帅蟲, 顯示040609_Ec_KT 纖毛帅蟲非純品系。此結果除提供檢驗寄生蟲純度的簡易方 法,也反映同一隻魚可被多種不同血清型之海水白點蟲混合感染,建議開發疫苗時須考慮多價或具交叉反應之纖毛帅蟲。綜合制動分析和ELISA 結果顯示海水白 點蟲至少存在四種不同血清型,其中以 111708_Cs_TT 所代表的血清型最顯著。 本研究更發現不同基因型的海水白點蟲,可能表現相同血清型。然而目前資料僅 能說明111708_Cs_TT亞洲主要血清型,無法得知海水白點蟲究竟存在多少種血 清型,需要進一步收集資料,做為開發多價疫苗之基礎。 | zh_TW |
