Title: 不同性質生質物和添加劑對複合型纖維酵素吸附及水解的影響

Abstract: 本研究利用不同的前處理產生不同性質的紙漿纖維,搭配複合型纖維酶來探討纖維素材料的物理及化學性質對酵素吸附水解的影響。利用纖維篩分不同大小纖維,和酸蒸爆、亞硫酸鹽製漿法及氧漂漂序來做不同木化性質材料。
在4oC低溫吸附結果顯示,纖維越小含較多比表面積有較好的可及性,全纖維素含量較多的全漂漿,則對複合型酵素有較大的吸附性,而在複合型和單一型酵素中,複合型酵素有較多極性區域可對不同性質的將有較大的親和性,對複合性酵素進而比較其酵素比活性,結果顯示木質素含量較高的酸蒸爆漿對雜蛋白有較高的親和性,而全纖維素含量較高的則對有效蛋白有較高的親和性,間切酶的吸附性又比木醣酶還要好。
在高溫水解中,利用產糖量和聚合度下降來對評估複合型纖維酵素的反應性,全漂漿可產較多的還原醣,聚合度也下降較快,而比表面積越大的小纖維產糖量無顯著增加,但可增加反應速率。另外利用不同酵素添加量,過多的添加在全纖維素含量較多的漿種可看出酵素反應中有脫附情形,產糖量和酵素添加量並無線性關係。
不同的蛋白質對不同漿種有不同的親和性,吸附性和反應性也會受影響,本研究添加BSA和Lysozyme,發現可有效的提升產糖量和水解速率,反應也較完全,木質素含量較多的漿種此趨勢越明顯。
最後分析其還原醣轉換率,發現化學組成為影響產糖量最主要因素,比表面積只影響其反應速率,且可利用添加不同添加劑影響速率和產率。
In this study, several attempts have been made to fabricate different properties of substrates by different pretreatments, and the correlation between properties of substrates and enzyme formulation was investigated.
At 4oC, the screening of pretreatment, the smaller fibers with more surface area have better accessibility, and C-tec have intimation with the bleached pulps which are fabricated by kraft cooking and process of DEDD. Relative to single enzyme, C-tec which is a kind of complex enzyme has affinity to substrates. Furthermore, the research demonstrated that lignin is intimate with irrelevant proteins and holocellulose take charge of effective proteins with measuring specific activity of supernatant. Moreover, the adsorption of CMCase is easier than Xylanase.
On the hydrolysis, more holocellulose have more accessibility and reactivity which are estimated by conversion of sugars and reduction of DP. The surface area can increase the efficiency, but the conversion is not different form the fiber sizes. On the other hand, with the enzyme loading increasing, the conversion of sugars is increased, but there is no linear relation between enzyme loading and yield. The desorption of phenomenon is observed with much enzyme loading。
According to the affinity of variation proteins, the accessibility and re were influenced by adding BSA and Lysozyme. That additives loaded can enhance the effective and efficiency, and the process can react completely. The more content of lignin correspond with the trend significantly. Finally, the analysis of conversion and DP can realize the effect of different substrates on hydrolysis.
