dc.description.abstractEB病毒(Epstein-Barr virus, EBV)隸屬於人類疱疹病毒科(Herpesviridae)的γ疱疹病毒亞科。EB病毒係第一個被報導與人類癌症直接相關的病毒,目前已知與多種惡性腫瘤及淋巴細胞增生疾病相關。EB病毒可將人類初代B細胞轉形成為具有無限增殖能力的的淋巴母芽細胞株(Lymphoblastoid cell line, LCL)。為了探討EB病毒將細胞不朽化的過程中對基因的調控,我們利用cDNA微陣列分析欲釐清在EB病毒感染後的基因表現。根據cDNA微陣列分析的結果,我們發現許多干擾素誘導基因(Interferon-stimulated genes, ISGs)在淋巴母芽細胞株中的表現量較在人類初代B細胞中高。由於干擾素誘導穿膜蛋白質(Interferon-induced transmembrane protein, IFITM)家族被報導具有抑制RNA病毒進入細胞或抑制RNA病毒複製的能力,因此我們以它們作為研究之焦點。而IFITM1的表現量為之中最高者,故挑選其作為本論文的研究目標。自即時定量聚合酶連鎖反應(real-time quantitative polymerase chain reaction, RT-Q-PCR)及西方墨點法(Western blotting, WB)的結果顯示,EB病毒潛伏期膜蛋白質1 (Latent membrane protein 1, LMP1)會利用其C-terminal activation region 1 (CTAR1)及CTAR2活化NF-κB的抑制劑進行處理後,IFITM1的表現量並未完全受到抑制,因此推測可能有其他因子亦可調控IFITM1的表現。以表現LMP1的細胞之條件培養液(conditional medium)培養Akata EBV(-)細胞的確觀察到IFITM1被誘導產生的現象。同時,我們亦透過反轉錄聚合酶連鎖反應(reverse transcription polymerase chain reaction, RT-PCR)觀察IFN-β在表現LMP1的細胞中的表現量上升。因此,我們推測干擾素可能參與在LMP1調控IFITM1表現的路徑當中。由目前的實驗結果顯示LMP1可能係透過直接及間接的方式調控IFITM1的表現。在探討IFITM1生物功能的方面,我們以慢病毒感染方式將淋巴母芽細胞株中的IFITM1剔除,發現IFITM1可協助維持淋巴母細胞株中的EB病毒處於潛伏期。zh_TW
