dc.description.abstract | 細胞主要利用泛素-蛋白酶體系統 (Ubiquitin-proteasome system) 和細胞自噬 (Autophagy) 兩機制將不需要的物質降解並維持體內恆定,而泛素 (Ubiquitin) 在其中扮演重要角色,除了協助兩者標記欲降解的物質,也可改變酵素活性或分子在細胞中的位置。目前對於泛素化 (Ubiquitination) 的了解較為透徹,且部份的泛素連接酶 (E3 ubiquitin ligase) 如 Parkin 與 Traf6 已被報導與細胞自噬有關。但泛素化的逆反應:去泛素化 (Deubiquitination) 及與細胞自噬的關聯性目前仍有相當大的研究空間。人體基因組中含有超過 90 種去泛素酶 (Deubiquitinating enzyme)負責進行此逆反應。根據其保守的催化域 (conserved catalytic domain) 可分為六群:USP, UCH, MJD, OUT, JAMM 和 MCPIP。最近研究指出 USP36 失去正常功能時會在營養充足的情況下促進細胞自噬的進行。藉由胺基酸序列的分析比對,我們在果蠅中找出超過 28 種去泛素酶,並進一步研究去泛素酶在調控細胞自噬中扮演的角色。除了 USP36,我們額外發現四種去泛素酶可促進或抑制細胞自噬的進行。我們的發現指出去泛素酶確實參與了細胞自噬的調控。 | zh_TW |
