dc.description.abstract | 奇異變形桿菌(Proteus mirabilis)是革蘭氏陰性的兼性厭氧的腸內菌,為健康人類腸道中的正常菌叢,但是在部分插尿導管的病患身上會造成伺機性感染,是尿道感染的重要病原菌,也會導致膀胱炎、腎結石,腎臟病等併發症。外膜是革蘭氏陰性菌特別且重要的組成,其位在細胞壁外層,為細菌與外界物質接觸的最外圍構造,其組成包含磷脂質、脂多醣和外膜蛋白;其中,外膜蛋白在細菌中扮演重要角色,除了為許多噬菌體的受器、小分子的通道等角色,還有細菌與宿主間傳遞訊息、保護細菌免於宿主攻擊等功能;外膜蛋白會感受外在環境的變化,改變自己的表現量去適應環境,許多外膜蛋白會在環境艱困時大量表現。在P. mirabilis中,OmpA和OmpF為不論任何狀況(宿主體外、營養物缺乏、宿主體內),都有一定大量的表現,為P. mirabilis非常主要的外膜蛋白;ompA在許多陰性菌中研究顯示為影響細菌感染、生存、適應環境有關,而ompF則與抗藥相關,但在P. mirabilis中還不甚清楚,本篇論文主要研究,ompA和ompF在P. mirabilis中的角色。
實驗設計為利用ompA和ompF的knockout,突變影響許多毒力因子和壓力耐受性,ompF突變株降低了表面移行能力、生物膜形成能力上升、改變許多抗生素的感受性和對抗酸、高滲透壓及氧化壓力的能力下降,也降低其入侵細胞的能力。ompF突變株在表現型上較多與野生株的差異,想了解在基因調控上ompF的調控路徑, RpoE和CpxR都是面對壓力環境下會活化調控下游基因的regulator,其活化條件都包括不成熟的外膜蛋白,兩者都可以自行調控,藉由ompF突變株發現rpoE和cpx的表現皆下降。為了瞭解ompF的基因調控,尋找本實驗室先前研究的regulator及根據ompF上游序列預測可能的regultor 以及可能的small RNA,找出一些可能調控ompF的基因,其中rybB利用real-time PCR證實其會抑制ompF的表現量,同時發現rpoE突變株ompF表現量也會上升。根據實驗室先前cytokine array的實驗結果,rpoE、rybB及hfq突變株中IL-8及MIF的量較野生株高,其共同點為OmpF表現量高,故假設OmpF會刺激細胞產生cytokine,並證實ompF突變株感染細胞後IL-8的產生量相對野生株下降很多。總結,ompF在Proteus mirabilis中扮演重要角色,影響移動性、毒性、抗藥性及與宿主的反應,並藉由rpoE和rybB調控ompF。 | zh_TW |
