| dc.description.abstract | 本論文研究目的在於利用不同濃度的菇蕈多醣體飼料,經由投餵的方式給予實驗白蝦並檢測白蝦其總血球數 (THC)、原酚氧化酵素活性 (proPO)、超氧陰離子的產生率(O2-)、吞噬率 (PR)、吞噬指數 (PI),以探討菇蕈多醣體給予對於白蝦先天性免疫反應的影響,同時在不同時間點予以病原體,觀察白蝦免疫力指標與抗溶藻弧菌能力的關係和存活率,以分析出適合的劑量及投餵頻率供現場養殖業者作為參考。結果顯示如下:
第一部分-菇蕈多醣體對白蝦免疫力的影響,三種不同的多醣濃度(1、2、4 g / kg diet) 對總血球數 (THC)、原酚氧化酵素活性 (proPO)、超氧陰離子的產生率(O2-)、吞噬率 (PR)、吞噬指數 (PI)等免疫指標均有顯著提升 (p<0.05) 的效果。尤其以高劑量組(4 g / kg diet)能最快提升免疫指標(THC、proPO、PR、PI),低劑量組(1 g / kg diet)需要較長的時間提升免疫力。 第二部分-菇蕈多醣體對白蝦感染溶藻弧菌後其免疫力和存活率的影響,實驗結果顯示,白蝦在接受溶藻弧菌試驗後,與Blank組(僅注射PBS未含細菌)相比,免疫指標均有明顯下降,高劑量組(4 g / kg diet)在溶藻弧菌感染實驗期間,能使proPO活性、O2-產生率、PR、PI、THC在短時間內提升,且達到最高。在抗菌能力方面,每組連續投餵後第0、1、3、5、7、10天進行溶藻弧菌攻毒(〖4.5×10〗^5 cfu shrimp-1),其48 h之存活率為40、80、70、60、70、80 %。低劑量組(1 g / kg diet)需要較長的時間提升免疫力,在抗菌能力方面,每組連續投餵後第0、1、3、5、7、10天進行溶藻弧菌攻毒(〖4.5×10〗^5 cfu shrimp-1),其48 h之存活率為40、70、50、50、60、60 %。Control組存活率為40、40、40、50、50、40 %。 綜合本研究結果發現,飼料中添加菇蕈多醣體,可有效提升白蝦先天性免疫反應與抗溶藻弧菌能力。 | zh_TW |
| dc.description.abstract | The purpose of this study is using feed with different concentrations of β-glucan on white shrimp, and examine the variance of total haemocyte count (THC), prophenoloxidase (proPO), superoxide anion (O2-), phagocytic rate (PR), phagocytic index (PI). It is investigate the effect of β-glucan on innate immune response of white shrimp and observed the immune parameter, survival rate of white shrimp which injected the pathogens at different fed time points. It can be analyze the appropriate dosage and frequency of feeding for the aquaculture industry.
Section I, different dietary doses (1、2、4 g / kg diet) of β-glucan were used. The result showed that the THC, proPO, O2- production ratio, PR and PI were higher significantly (P<0.05) than the control group. The high-dose group (4 g / kg diet) can enhance the immune parameters fastest (THC, proPO, PR, PI). The low-dose group (1 g / kg diet) take a more time to increase immune parameters. Section II, white shrimp after injected the Vibrio alginolyticus compared with the Blank group, the immune parameters were significantly reduced. The high-dose group (4 g / kg diet) can enhance the immune parameters fastest, and the survival rate of white shrimp which injected V. alginolyticus after 48h at different fed time points is 40、80、70、60、70、80 % (day 0、1、3、5、7、10). The low-dose group (1 g / kg diet) take a more time to increase immune parameters, and the survival rate of white shrimp which injected Vibrio alginolyticus after 48h at different fed time points is 40、70、50、50、60、60 % (day 0、1、3、5、7、10).
