dc.description.abstract我們用大小均一、排列整齊的明膠泡泡製造細胞鷹架,並在其中養3T3纖維母細胞。我們對細胞如何對應其外微環境協調其細胞骨架感興趣。我們利用微流體通道的特性製造一系列尺寸不同孔徑但同孔隙率的鷹架,種進3T3纖維母細胞後用Phalloidin 染細胞內部的肌動蛋白,利用共軛焦顯微術取得細胞影像。用Imaris重建三維影像,針對細胞內三個軸向長度做統計.zh_TW
dc.description.abstractWe have used mono-dispersed and self-packing gelatin bubbles for our scaffolds. 3T3 fibroblasts were used for our cell model. We were interested in how cells responded
to their environment providing different geometries and mechanical properties such as surface curvature and stiffness. We utilized soft lithography to construct our scaffolds of different pore sizes and gas fractions. We cultured 3T3 in the scaffolds, stained their cytoskeletons with Phalloidin and took images with confocal microscopy. We reconstructed cell images using Imaris and took measurements of cells in scaffolds.
