| dc.description.abstract | 黃麴毒素為食品與飼料中常見的黴菌毒素。當牛隻食用含黃麴毒素B1的飼料後,其乳汁中可能產生代謝物黃麴毒素M1,具有肝毒性與致癌性。因為一般消毒過程無法去除黃麴毒素,各國莫不對其加強管制。歐盟自1999年起將黃麴毒素M1在鮮乳中的容許量由0.5 μg/L降至0.05 μg/L, 對現有檢測方法形成挑戰。
分析鮮乳或奶粉中黃麴毒素M1的確認方法,以液相層析儀及螢光偵測為主;因需要經由衍生化來增強螢光性,過程耗時且使用毒性試劑。樣本前處理則常以固相萃取加上免疫親和性管柱淨化;因管柱流速緩慢,僅使用20-50毫升的樣本,限制了靈敏度,再加以無適當的內標準品,導致分析變異性頗大。 本研究改善分析靈敏度,並縮短檢測時間。在樣本的萃取、淨化及儀器分析條件的最適化做了以下的探討︰(1)以質譜/質譜儀取代螢光偵測;(2)以圓盤型吸附劑增大樣本體積;(3)樣本淨化方法之比較;(4)嘗試合成氘或氧18標定的穩定內標品;(5)分析方法驗證。 本研究顯示牛奶樣本可大至200毫升而無破出情形,且較一般檢測方法樣本加大了4∼10倍。分析方法之精密度與回收率,隨著不同基質及不同淨化方式而有所t異︰(1)以免疫親和性管柱淨化:全脂奶粉、全脂鮮奶以及低脂鮮奶的回收率分別為:67.9 ± 9.3%、78.2 ± 7.3% 及86.6 ± 4.1%(平均值 ± 標準差);方法偵測極限全脂鮮奶及低脂鮮奶各為0.59 ± 0.19 ng/L(1.02 ng/L)及0.66 ± 0.12 ng/L(0.94 ng/L)(平均值 ± 標準差(99% 信度方法偵測極限))。(2)以多功能淨化管柱淨化:全脂鮮奶及低脂鮮奶的回收率分別為6.51 ± 0.28% 及15.8 ± 4.6%(平均值 ± 標準差);方法偵測極限各為14.1 ± 2.6 ng/L(24.6 ng/L)及9.22 ± 3.34 ng/L(16.8 ng/L)(平均值 ± 標準差(99% 信度方法偵測極限))。以免疫親和性管柱及多功能淨化管柱,分別淨化認証參考樣本BCR-282奶粉,方法偵測極限分別為0.0085 ± 0.00050 μg/kg(0.0096 μg/kg)及0.0082 ± 0.0022 μg/kg (0.013 μg/kg)(平均值 ± 標準差(99% 信度方法偵測極限))。 內標品的合成上,本研究顯示確有氧16與氧18交換現象,但反應無法趨於完全,未反應之16O-黃麴毒素將造成偽陽性;以氘標定的方式,因反應條件需在鹼性、高溫及長時間進行,使得黃麴毒素分解,無法合成穩定的同位素標定內標品。而以黃麴毒素B1作為回收標準品,其訊號強弱與M1之趨勢並不一致,效果也不盡理想,並不適合做為內標品。尋找一個適合定量牛奶中黃麴毒素M1之內標品,仍有待進一步的研究。 本方法應用於分析鮮奶,方法偵測極限比現存方法低了三至五百倍,為牛乳檢測提供優異的靈敏度。 | zh_TW |
| dc.description.abstract | Milk may contain aflatoxin M1 (AFM1) if cows ingest aflatoxin B1(AFB1), which is a usual contaminant in animal feed. AFM1 is toxic and carcinogenic to liver, and can survive after pasteurization. The European Union has reduced the regulated level down to 0.05 μg/L and has challenged the existing analytical methods.
To confirm the existence of aflatoxin M1, the most used measurement is by high-performance liquid chromatography (HPLC)/fluorescence detection.
