dc.description.abstract本文是以原子力顯微鏡探討單一生物分子之間作用力為主,內容分成一量測Anti-Human IgG1與Human IgG1之間的解離力,探討解離力與壓入深度以及停留時間的關係,比較下壓時間與停留時間對解離力的影響,從數據中知道到了停留時間十秒時仍未有穩定的解離力出現,可見構形仍然在持續改變著。另外研究蛋白質拉伸的實驗,實驗樣本為丙胺酸(poly-alanine),探討去摺疊力與實驗環境酸鹼度以及停留時間的關係,並將實驗所得的力-距離曲線與Worm Like Chain model做結合,如此可以進而求得丙胺酸的抗彎長度(persistence length)與實驗環境酸鹼度的關係,並與圓旋光光譜儀的實驗結果最比較,實驗結果證實不同酸鹼溶液下去摺疊力的大小與該胺基酸的pKa值息息相關。zh_TW
