dc.description.abstract | 台灣四面環海,海洋生物資源仍具有相當大的開發潛力,前人研究多在高溫環境分離耐熱細菌,本研究從台灣沿岸海水中篩選能生長在45 oC環境下的耐熱細菌,共分離出88株耐熱細菌,根據16S rDNA片段之RFLP (DdeI、HhaI、RsaI)基因型分析及革蘭氏反應、溫度、鹽度等特性將此88株細菌分成12群。分離株多為革蘭氏陰性菌,可生長在含0.5-12.5 % NaCl的PY培養液中,生長溫度範圍在4-50 oC之間。根據分離株的16S rDNA片段序列與已知菌株比對後,12群分離株分別為Alcanivorax、Bacillus、Exiguobacterium、Idiomarina、Marinobacter、Marinomonas、Mesorhizobium、Nitratireducter、Pseudidiomarina、Salinivibrio、Vibrio等屬,研究結果顯示出台灣沿岸海水中具有豐富的耐熱菌菌相,而且多分布在γ- Proteobacteria綱。其中分離株E8的16S rDNA序列與最相近的菌種Idiomarina fontislapidosi、Pseudidiomarina tainanensis相似度分別只有92.5 %與94.0 %,可能為新種細菌。分離株E8為革蘭氏陰性細菌、為具有單一極鞭毛之桿菌,生長溫度範圍為4-45 oC,最適合生長溫度為37 oC,可生長酸鹼值範圍為pH 7-9,可生長鹽度範圍為0.5-10 % NaCl,具有尿素酶、解脂酶、明膠酶、蛋白酶活性,DNA G+C含量為51.5 mol %,於37 oC培養時細胞脂肪酸組成大於5 %者有iso-C15:0 (24.83 %)、iso-C17:0 (23.85 %)、iso-C17:1 ω9c (16.32 %)、C16:0 (9.95 %)、iso-C11:0 3-OH (5.35%),根據形態、生理、生化及遺傳特性分析,E8應為Idiomarinaceae科的新穎細菌。 | zh_TW |
dc.description.abstract | Taiwan is surrounded by sea, the marine biological resources nearby still has development potentials. Past researches focused on the isolation of thermotolerant bacteria from the high temperature environments, but this study aims to isolate thermotolerant bacteria from coastal seawater with normal temperature. This study selected thermotoletant bacteria which can grow at 45 oC from Taiwan coastal seawater and is divided into 88 isolates. According to RFLP genotypes (DdeI、HhaI、RsaI) of 16S rDNA fragments, Gram test, growth temperature range and growth salinity range, divided these 88 bacteria into 12 groups. These isolates are mostly Gram negative bacteria, and they can grow in PY broth that contains 0.5-12.5 % NaCl with a temperature range from 4-50 oC. According to the comparison between 16S rDNA sequence with known bacteria, these 12 groups of bacteria can be defined as Alcanivorax、Bacillus、Exiguobacterium、Idiomarina、Marinobacter、Marinomonas、Mesorhizobium、Nitratireducter、Pseudidiomarina、Salinivibrio、Vibrio. This result indicates that there are abundant thermotolerant bacteria in the coastal seawater of Taiwan and the γ- Proteobacteria is the major group. One of these isolates, E8, its 16S rDNA sequence is similar to Idiomarina fontislapidosi、Pseudidiomarina tainanensis with similarity of 92.5 % and 94.0 %. Isolate E8 is a Gram negative rod bacterium with monotrichous flagellum, it can grow at 4-45 oC, pH 7-9, 0.5-10 % NaCl in PY broth, and with optimal growth temperature at 37 oC. E8 contains urease, lipase, gletinase, protease activity. The DNA G+C content of E8 is 51.5 mol%. At 37 oC, the cellular fatty acid contents of E8 larger than 5 % are iso-C15:0 (24.83 %)、iso-C17:0 (23.85 %)、iso-C17:1 ω9c (16.32 %)、C16:0 (9.95 %)、iso-C11:0 3-OH (5.35%). According to cell morphology, physiology, biochemistry and genetic characters, isolate E8 should be a new bacterium of Idiomarinaceae. | en |
