| Abstract: | 本研究使用父、母、染病孩子三元體的資料結構,分別利用統合參數概似函數及傳遞不平衡檢定,來進行連鎖分析。考慮在需要使用逐次過程的情況下,給定特定的疾病基因頻率在不同的遺傳模式下,根據本研究模擬分析顯示,使用結合統合參數概似函數和逐次機率比例檢定(SPRT)的統計量會比結合傳遞不平衡檢定(TDT)和逐次機率比例檢定的統計量到達判定界限所需的樣本數少。 In this paper, we use case-parent triad families as the data structure to demonstrate how to proceed sequential linkage analysis. We adopt two types of linkage methods, the unified-parameter likelihood method and the transmission/disequilibrium test (TDT) method, to implement this sequential test procedure. Under different genetic scenarios, our simulation studies show that the unified-parameter sequential likelihood method requires less sample sizes to reach the sequential decision boundary than the TDT sequential method. |
