摘要: | 本研究探討菇蕈多醣體(mushroom glucan)對於小鼠免疫反應以及移殖瘤的影響,實驗分兩部分:
第一部份以胃管灌食或腹腔注射的方式給予多醣體,持續四周,測量小鼠身體指數、吞噬作用、自然殺手細胞細胞毒殺活性、以及血液中細胞素的含量。結果顯示不論是口服或腹腔注射多醣體皆不影響小鼠的體重、肝體比或腎體比。血液中的顆粒球細胞(granulocyte)與單核球細胞(monocyte),不論是在表現吞噬的百分比或噬入外來物數量上均比對照組高且有顯著差異 (P<0.05);自然殺手細胞細胞毒殺活性方面,口服或腹腔注射多醣體的實驗組在第二週以後的測量值高於對照組且達到顯著差異(P<0.05);血液中細胞素的含量,口服或腹腔注射多醣體皆觀察到TNF-α(tumor necrosis factor-α)、INF-γ(interferon-γ)的含量高於控制組。由以上的結果可知,經口投餵或腹腔注射菇蕈多醣體能活化小鼠的免疫系統。 第二部分將小鼠殖入Lewis lung carcinoma cells,以胃管灌食或腹腔注射的方式給予多醣體,觀察多醣體的給予是否能抑制腫瘤的生長。結果發現,多醣體實驗組的腫瘤重量平均值皆較對照組小,口服多醣體組與對照組有顯著差異(P<0.05);口服多醣體組的腫瘤抑制率為40.58%,腹腔注射多醣體組為21.01%。 This research was to study the mushroom glucan’s immunomodulatory and antitumor effects in vivo. In the first part, mice were treated with glucan by gavage or intraperitoneal daily. Administration of glucan for four weeks didn’t affect the growth and relative organ weight of Balb/c mice. The phagocytosis activities of monocytes and granulocytes in blood were significantly increased. After one week, the cytotoxicity of natural killer cells was higher than the result of control group and reached significantly different. The experimental groups were detected higher level of tumor necrosis factor-α and interferon-γ in blood than the PBS group. Oral or intraperitoneal administration of gluacn can improve the immune system of mice. In the seconed part, mice were implanted Lewis lung carcinoma cells subcutaneously. The group that oral administration of glucan for twelve days had smallest solid tumors, while control group had biggest tumor, i.p group among these two groups. The tumor inhibition rate of oral glucan group was 40.58%, and i.p. glucan group was 21.01%. |
