dc.description.abstract | 生物材料常被添加於自體脂肪移植體,既可作為所移植之脂肪細胞的支撐材料,又可作為脂肪衍生幹細胞(Adipose-derived stem cells,ADSCs)的細胞載體。本項研究選用海藻酸(Alginate)作為自體脂肪移植之支架,由於其細胞不易貼附於海藻酸之特性,將海藻酸結合層黏連蛋白(Laminin)修飾成較親細胞之層黏連蛋白-海藻膠微球(Laminin-Alginate Beads),進一步研究使用自體脂肪移植體模型,在免疫功能正常的大鼠對層黏連蛋白-海藻膠微球、脂肪細胞(Adipocyte)及脂肪衍生幹細胞進行檢測。
此研究中我們將不同組合的切碎之自體脂肪組織(稱為「A」,代表脂肪組織)、層黏連蛋白-海藻膠微球(稱為「B」,代表微球)及脂肪衍生幹細胞(稱為「C」,代表細胞)植入15隻 Sprague-Dawley大鼠的背部。A組僅植入脂肪組織,B組僅植入層黏連蛋白-海藻膠微球,AB組植入脂肪組織加上層黏連蛋白-海藻膠微球,BC組植入包覆脂肪衍生幹細胞的層黏連蛋白-海藻膠微球,ABC組植入脂肪組織及包覆脂肪衍生幹細胞的層黏連蛋白-海藻膠微球。在植入後第1、6及12週時,以7T動物用核磁共振影像儀(7- tesla Magnetic Resonance Imaging)評量植入物於大鼠體內的狀況。第12週時,犧牲大鼠,取出植入的材料進行肉眼檢查及組織學評估。 核磁共振影像(Magnetic Resonance Imaging,MRI)、肉眼檢查及組織學資料的結果皆顯示,ABC組植入物有較佳的生物材料再吸收作用,且移植之脂肪組織存活狀況及脂肪衍生幹細胞之成脂分化作用皆獲得改善。ABC組的移植體體積保留量(89%)亦極顯著高於A組(58%)(p<0.01)。我們的發現顯示,併用切碎的脂肪組織與包覆於層黏連蛋白-海藻膠微球的脂肪衍生幹細胞可得到較佳的自體脂肪移植脂肪結果。 | zh_TW |
dc.description.abstract | Biomaterials are often added to autologous fat grafts both as supporting matrices for the grafted adipocytes and as cell carrier for adipose-derived stem cells (ADSCs). In this study, a new materials were synthesized by linking developmentally essential ECM and laminin to alginate(Alg). The production of new material was followed by an in vivo study, which used an autologous fat graft model to test the laminin-alginate biomaterial, adipocytes, and ADSCs in immune-competent rats.
In this study, we transplanted different combinations of shredded autologous adipose tissue [designated “A” for adipose tissue]), laminin-alginate beads [designated “B” for bead], and ADSCs [designated “C” for cell]) into the backs of 15 Sprague-Dawley rats. Group A received only adipocytes, Group B received only laminin-alginate beads, Group AB received adipocytes mixed with laminin-alginate beads, Group BC received laminin-alginate beads encapsulating ADSCs, and Group ABC received adipocytes and laminin-alginate beads containing ADSCs. Seven-tesla magnetic resonance imaging was used to evaluate the rats at the 1st, 6th, and 12th weeks after transplantation. At the 12th week, the rats were sacrificed and the implanted materials were retrieved for gross examination and histological evaluation. The results based on MRI, gross evaluation, and histological data all showed that implants in Group ABC had better resorption of the biomaterial, improved survival of the grafted adipocytes, and adipogenic differentiation of ADSCs. Volume retention of grafts in Group ABC (89%) was also significantly greater than those in Group A (58%) (p<0.01). Our findings support that the combination of shredded adipose tissue with ADSCs in laminin-alginate beads provided the best overall outcome. | en |
