摘要: | 香水文心蘭(Oncidium Sharry Baby)是台灣花卉市場上一個很受歡迎的栽培種,它會開帶有巧克力香氣的紫色花朵,並且香氣的散播會隨著一日的不同時間而改變。而在花卉市場上的另一個栽培種,南西文心蘭(Oncidesa Gower Ramsey),則不帶有香氣。花的香味對植物來說有重要的功用,如抗蟲咬(antifeedant)、抗微生物(antimicroorganism)等等,而最重要的是香味可以吸引授粉者前來授粉(pollination),以達到植物繁衍下一代的目的。自然界中花的香味則可以分成三類的化合物:萜類化合物(terpenoids)、苯類/苯丙烷類(benzoids/ phenylpropanoids)化合物、以及脂肪酸的衍生物(Fatty acid derivatives)。
在本研究中,我們想要了解在香水文心蘭中主要的香氣成份分子,以及產生這些香味分子的基因,並進一步研究是何種基因表現及調控機制的差異造成香水文心蘭及南西文心蘭產生香味與否。在本文的研究之中,我們先藉由氣相層析-質譜技術(GC-MS),分析出在香水文心蘭的主要香氣成分為單萜類(monoterpenoids)的羅勒烯(β-ocimene)及芳樟醇(linalool),並且發現它們在香水文心蘭中白天時散發的量比較多,而在下午或晚上有逐漸減少的情形,並進一步找出可能為主要生合成酵素的萜類合成酶(terpene synthase (TPS))。在香水文心蘭中,萜類合成酶受到CCA1(circadian clock associated 1)轉錄因子的調控,故其基因表達會具有概日韻律(diurnal rhythm)。在分析兩種文心蘭萜類合成酶的啟動子(promoter)序列時,我們發現在香水文心蘭中萜類合成酶的啟動子中,具有一個南西文心蘭缺乏的順式作用元件(cis-element)—CCA1結合位點(CCA1 binding site (CBS), 5’-AGATTTTT-3’),使其得以和CCA1做結合而進行萜類合成酶的表達。 本研究提供了在文心蘭中香氣成分分析的基礎,希望在此基礎之上,將來能有更成熟的香氣轉殖株產生,為台灣的文心蘭花卉市場增添新的競爭力。 Oncidium Sharry Baby (Onc. Sharry Baby) is a kind of popular cultivar orchids in horticulture. It blooms purple flowers with a sweet chocolate fragrance, which was emitted differentially with a circadian rhythm, while Oncidesa Gower Ramsey is not with the similar floral scent. In general, floral scents have important capabilities of antifeedance, antimicroorganism and attracting pollinator. Floral scent compounds are classified into three types: terpenoids, benzenoids/phenylpropenoids, and fatty acids. In this study, we attempt to analyze the diversity compounds of floral scent, and further investigate the differential gene expressions and regulatory mechanisms between aromatic Sharry Baby plant and non-aromatic Gower Ramsey plant. Here, gas chromatography-mass spectrometry (GC-MS) was applied to analyze the amount of floral scent compounds in Sharry Baby and Gower Ramsey plants. The result revealed that the monoterpenoids, β-ocimene and linalool, are more abundant in Sharry Baby than in Gower Ramsey. Besides, the accumulations of β-ocimene and linalool were peaked at day and low in night in Sharry Baby plants. Next, one of the key genes in monoterpene biosynthesis pathway was identified as terpene synthase (TPS), whose expression patterns were oscillated, similar with the floral scent emissions in Sharry Baby. Furthermore, the mechanism of circadian emission of floral scents in Onc. Sharry Baby is controlled by the oscillator gene, CCA1(CIRCADIAN CLOCK ASSOCIATED 1) under light condition. CCA1 transcription factor increases thee expression level of terpene synthase (TPS) by binding on CBS motif, 5’-AGATTTTT-3’, of promoter region to affect the circadian emission of floral scents in Onc. Sharry Baby. We expect that more transgenic Oncidium lines with lovely fragrance can be generated in the near future. |
