摘要: | 在本篇論文中,主要目的為觀察雞胚胎角膜發育之情形。以萊亨雞為動物實驗對象,使用不同天數(十至十九天)之雞胚胎,以H.H. Stage輔助判斷其孵育時間,每二十四小時來作為角膜階段性成熟的劃分,分別取下左眼及右眼角膜,每一時期以三組為單位,取樣後使用解剖顯微鏡來觀察不同天數的胚胎中角膜尺寸之發展以及使用光譜儀定量其透光率,追蹤變化趨勢,並以高解析度白光顯微鏡分析角膜隨著孵育時間不同而產生的圖樣以及結構變化,其中主要觀察部分為角膜內膠原蛋白纖維之排列與生成情形,除了比較不同成熟度之角膜結構外,同時也比較了尺寸以及穿透率之差異。經由不同發育階段雞胚胎中的角膜觀察及量測,可以推斷角膜生成之情形以及內部構造變化,並追蹤角膜內膠原蛋白纖維排列結構與生成模式,使眼科學上對於雞胚胎之角膜發育有更進一步之發現,同時也能與更多角膜生成相關研究比對,建構出更可靠雞胚胎角膜之發育模型。 The optical microscope has been developed for many centuries of science, first with the invention and use of lenses, and then with the perfection of lenses and illumination. Today, due to the integrity, fineness and nondestructive of measurement, optical microscopy has been widely used in the biological and medical imaging. Cornea, the outermost layer of the eye, is mainly composed of fibrotic collagen. Any distortion of cornea collagen orientation can result in significant visual deterioration. In order to understand how corneal collagen is organized and the probability of fabricating the artificial cornea to cure corneal diseases. It is an important work to realize the development of cornea. In this thesis, the main purpose is to obtain the development of chicken embryo cornea. Leghorn species was chosen as the experiment animal, for observing the collagen pattern and measuring the size, also the transparency of cornea. We first utilized high resolution optical microscopy to show the detail of structure. Then we used the dissecting microscope and Vernier caliper to evaluate the dimensions of cornea during growth. At last, the spectrometer was used to study the transparency in the wavelength range of 200 nm -1000 nm. The observation period of chicken embryo cornea development started from 10 to 19 day of post fertilization. Three embryos were used to support the reliability of the experiment for each day of post fertilization. The result of this study not only shows the growth details of cornea, but also provides the valuable information for us to inference the internal structure of chicken embryo. |
