dc.description.abstract | 本實驗乃擬嘗試利用各種不同之培養基,來培養廣東住血線蟲(Angiostrongylus cantonensis )之第一期幼蟲,使之不僅能存活於寄主體外,且能蛻皮或發育。 廣東住血線蟲之第一期幼蟲,自老鼠糞便中以Baermann的方法分離出來後,於無菌操作箱中,以含5?“Chloros”之Hanks氏平衡鹽液(HBSS)及Antibiotics-antimycotic混合液雙重消毒後,培養於RPKI-1640 medium,Macoy's medium,Mimimum Essential Medium (MEM)及Medium 199中,幼蟲皆可存活約27-29天,而以M199之效果最好,又分別於MEM及M199培養液中加入10?40%之Foetal Calf Serum (FCS)或Newborn Calf Serum (NCS),以及1% Yeast extract,1% Vitamin與10% Snail extract等添加物。實驗結果顯示,這些物質對延長幼蟲之存活日數,或協助幼蟲蛻皮方面,並無加強作用。在以上幾種培養液中,蟲體被培養10-13天時,尚約有50%以上之蟲體保持在活動的狀態,但當蟲體培養於自來水中時,則第一期幼蟲約僅存活三天,而於12小時後,已有50%以上之蟲體降低活動力。 幼蟲感染於水生螺絲Biomphalaria globrata後,蟲體之全長及最大寬度均有顯著的增加,在感染的第七天及第十七天分別有兩次蛻皮現象發生。於感染後第十八天,體寬增加約一倍,體長則增加約34%。當幼蟲被培養於MEM中15天後,蟲體之最大寬度增加約13%,而體長則未有增加之現象。 廣東住血線蟲第一期幼蟲之體外培養,最適當的PH值?6.0-8.0之間,至於溫度之影響,則在37℃下幼蟲之活動力較高,而於26℃之活動力較差,但此兩種不同溫度下幼蟲之存活日數皆約28天。 本研究所得之結果,雖不能使細蟲蛻皮或進一步之發育,但蟲體約有一個月的存活期,且50%之幼蟲可在10?13天之培養期間內維持高度的活動能力,已足以供給本幼蟲之生理,生化或藥物試驗之用。 | zh_TW |
