dc.description.abstract | 腦下垂體所分泌的促性腺激素(gonadotrophin)對於卵巢之類固醇生成作用(steroidogenesis)的調控已大致被研究清楚,近年來又發現一些卵巢本身分泌的物質,具有autocrine或paracrine性質,而對於類固醇生成作用的調控亦有貢獻。本實驗的目的在於探討二十碳三烯酸(dihomo γ-Linolenate)與二十碳四烯酸(arachidonate)及其代謝物對於泥鰍的卵巢卵細胞之類固醇生成作用的影響。低濃度的arachidonic acid(25-200μM)對於睪丸酮的生成有促進,但不影響hCG(10IU/mL)所誘導的睪丸酮生成,高濃度的arachidonic acid(400?1200μM)則促進作用漸減,且抑制hCG所誘導的睪丸酮生成。dihomo γ-Linolenate則只有在濃度50μM時對於睪丸酮生成有微小但顯著之促進作用,其餘濃度(25μM及100-1600μM)均無顯著作用,但在高濃度(100?800μM)下則對於hCG所誘導的睪丸酮生成有抑制作用。 前列腺素El(PGE1)及前列腺素E2(PGE2)在各濃度下對於睪丸酮生成均有顯著的促進作用,4μM濃度的PGE1及PGE2其促進睪丸酮生成的效果便與hCG(10IU/mL)相當,而前列腺素對於hCG(10IU/mL)所誘導的睪丸酮生成具有協同作用。前列腺素Flα (PGF1α)及前列腺素F2α(PGF2α)對於睪丸酮生成有些許促進效果,但不呈現劑量相關,且對於hCG所誘導的睪丸酮生成沒有明顯的促進或抑制效果。INDO(cyclooxygenase抑制物)幾乎完全抑制arachidonic acid(100μM)對於睪丸酮生成的促進作用, NDGA(lipoxygenase抑制物)亦有些許抑制效果,但效果遠不如同濃度的INDO,顯示arachidonic acid的促進效果主要是藉由轉變成cyclooxygenase代謝產物而達成;另外,INDO亦抑制hCG所誘導的睪丸酮生成,顯示cyclooxygenase代謝產物參與hCG 所誘導的類固醇生成作用。PGE1(8μM),PGE2(8μM)及arachidonic acid(100μM)對於睪丸酮生成的促進作用均因培養基中IBMX(cAMP phosphodiesterase抑制物)的存在而明顯增強,顯示此三者的作用,至少部分以cAMP為其次級訊息傳遞物質;PGE1(8μM) ,PGE2(8μM)及arachidonic acid(100μM)刺激睪丸酮生成作用隨時間變化情況極為類似,且均再加強25-hydroxycholesterol(5ug/mL) 所誘發的睪丸酮生成,顯示三者極可能經由相同的作用機制來達到促進類固醇生成作用的效果。 本實驗為首度證實前列腺線對於魚類卵巢之類固醇生成具有調控作用,至於更詳細的調控機制,則有待更進一步深入探討。 | zh_TW |
