| dc.description.abstract | 電穿孔法(Electroporation)常被用於胚胎與細胞株之基因或其他物質轉殖之用,但其穿孔機制與電脈衝擊之關係至今仍不十分清楚。本文嘗試以兩種不同電脈衝波型:1.指數式遞減波(Exponential decay wave)及2.方形波(Square wave)之電穿孔儀,以染劑伊紅為轉殖物質,藉以探討牡蠣卵在各種電場下,電脈衝擊之各參數值在物質轉殖上所扮演的角色,及不同卵割時期之牡蠣胚胎對電脈衝擊的耐受性與封孔效率之影響。本結果發現在指數式遞減波組中,高電容(125?960μF)與適當電壓(100?400V)的組合較適合以牡蠣卵進行物質轉殖,其所得電穿孔效率(ε % )為: C1.4•E2.35 /(1.671*10^9) ε%={1-(1/2) }•100% ε:電穿孔效率 C:電容 電容 E :電場強度 而在方形波組中,脈寬(Pulse width)、脈衝數(Number of pulse)及週期次數(Cycle)與物質轉殖有正相關性;而樣品體積(Volume) 及極針和液面距離(Dial)與物質轉殖間有負相關性。此外由反應時間(Burst time)可推測牡蠣卵封孔時間約為200?250msec.。在不同卵裂階段中,其卵隨不同卵裂時期對電脈衝擊的耐受性有增強的趨勢;而在指數式遞減波組中,其卵封孔時間約為20秒。應用上述實驗所得電穿孔法基本參數值特性,利用pmiwZ(β-galactosidase gene)為轉殖基因,以牡蠣、文蛤與豐年蝦為基因轉殖模式動物,經β-galactosidase活性測試發現,在控制組中,牡蠣、文蛤與豐年蝦皆僅在消化道呈藍色反應;在牡蠣與文蛤基因轉殖組中,其面盤幼蟲呈全身性藍色反應,少部分呈鑲嵌狀藍色反應。在2?16細胞期之牡蠣與文蛤胚胎進行基因轉殖時,藍色反應百分率各為5.36%±1.74%與19.12%±8.97%,在豐年蝦基因轉殖組中,其40隻無節幼蟲中,僅3隻在泳肢上呈藍色反應,而在豐年蝦子代測試中,第8與第37對豐年蝦子代呈全身性藍色反應,藍色反應百分率為4.44%±2.56%。 | zh_TW |
