dc.description.abstract | 將草蝦病原菌Vibrio vulnificus菌株TG617經0.3%福馬林去活性,製作抗原,免疫Balb/c老鼠。利用細胞融合瘤技術及ELISA篩選出13株融合瘤細胞,其中二株融合瘤細胞C3D5,C1E6所分泌之單株抗體可與異種弧菌例如V. damsela,V. harveyi,V. tubiashi及V. parahemolyticus作用,顯示V. vulnificus與其他弧菌間具有共同得抗原基團(epitopes)。其餘11株融合瘤所分泌之單株抗體則只特異地對V. vulnificus TG617菌株作用,但沒有任何一個單株抗體能同時辨認受測之13株V. vulnificus菌株,顯示各V. vulnificus菌株間之抗原組成具異型性(heterogeneous)。同時分析13株融合瘤分泌之單株抗體分析之結果發現,相同生物群(Biogroup)種,各菌株抗原基團不同,甚至同種寄主菌株間抗原基團亦不同,顯示受測之13株V. vulnificus菌株之抗原基團輿生物群或寄主種別間無關連,亦輿地區性無關。在養殖現場鑑定菌種時?避免漏檢,建議將融合瘤C3D5,C1E6分泌之單株抗體稀釋,降低C3D5,C1E6與其他弧菌間的交叉反應,再輿D3A6,D3D3及C1C2等3個融合瘤分泌之單株抗體混合,以做?診斷V. vulnificus病原菌之試劑。 至於海水養殖鰻所分離之菌株UE516迄今尚未篩選出可作用之單株抗體,須再篩選更多之融合瘤以找到可認識所有菌株之單株抗體。篩選出來之單株抗體除C2B2輿D3A11屬於IgG2b,C3D5屬於IgG3外,其餘均?IgG1。比較2個單株抗體是否競爭同一個抗原基團,發現C1F5及C3E4再ELISA呈色上並無加成性,因此可能?相同的單株抗體,也可能二者辨識之抗原基團其立體結搆(configuration)相似或反應位置(reaction site)相當靠近。 | zh_TW |
