dc.description.abstract | 本研究係利用虱目魚(Chonos chanos)的心臟組織,建立一連續細胞株(MH) ,此細胞株的形態呈表皮細胞狀,在含15%胎牛血清的L-15(Leibovitz's 15)培養液中,可良好的生長;細胞數目增加一倍的時間約二天.自民國七十七年九月至民國七十八年六月止,共經繼代培養達七十七代. 此細胞的生長隨血清濃度的增加而更加迅速;其生長的最佳環境為溫度28℃-31℃.pH8.0,8.5;滲透壓400-530mmol/kg .對於培養液的選擇,以MEM和L-15最好,此細胞株表面附著效應只有0.2%,但在使用條件性培養液時,可使附著效應上升至1%以上.分析第7,30,50代分裂中期細胞的染色體數目,顯示其分佈在26至42之間,而以34個染色體最多,藉電子顯微鏡的觀察,可見細胞的表面呈平滑狀,但在展開後有纖維產生,細胞株對於IPNV-Ab,IPNV-VR299,IPNV-Sp,EVE,EVA,EVEX及CV皆有感受性,但對於病毒的複製量卻不高.細胞經污染檢定後,沒有細菌,黴菌及黴漿菌的污染. | zh_TW |
