dc.description.abstract | 以東方果實蠅(Dacus dorsalis Hendel)成蟲之精果?材料,經過醋酸胴脂紅染色製成壓片。觀察壓片與經由紫木蘇,伊紅染色的石臘切片,均可發現精原細胞?包囊所圍繞,囊內的精原細胞形態相似,且其細胞週期亦同步化而細胞數目則?2(上標 n)(n?1至6之自然數)。推測每一完整之囊內係由一個精原細胞經6次同步分裂而成64個細胞的精原細胞群,這64個精原細胞肥大而轉化?64個初級精母細胞,兩次減數分裂仍在包囊中進行,囊中最後256個精子。 活體試驗中,以含氚的胸嘧啶核?(H3-thymidine)注射入羽化一、兩日的成蟲,然後在不同時期固定其精巢,並作成自動放射顯影之壓製片,以估計精母細胞減數分裂,精子成熟過程中所需之時日。結果發現:由減數分裂前精母細胞最後一次DNA合成期至第一次減數分裂中期約需6.50日;第7日可以發現少量的早期精細胞有銀粒標記。而由減數分裂前合成末期後第11日可見到少數的精子被標記,大量成熟的精子,在第13日出現銀粒。其中第一次減數分前期佔了相當長的時間,完成兩次減數分裂約需0.5-1日,精細胞變形?精子約需4日。 | zh_TW |
