摘要: | 鐵於生物體中扮演著重要角色,在哺乳動物血清中,輸鐵蛋白(transferrin)主要功能在輸送鐵至不同組織。而transferrin再藉著細胞膜上輸鐵蛋白受體(transferrin receptor)將鐵傳送至細胞膜內以供使用。 本實驗在製備transferrin抗體,並耦合horseradish peroxidase(HRP)至抗體上,以供酵素免疫分析之用,以三明治法酵素免疫分析所建系統,較佳測試範圍為10ng/ml-lug/ml,並分析各器官(腦,肺,心,肝,胰,脾,腎,睪丸,副睪,貯精囊,攝護腺,卵巢,子宮)中transferrin分佈情形,發現所測器官中皆有transferrin存在,而貯精囊中有極高量存在;另以transferrin抗血清試圖純化可能為transferrin receptor之蛋白質。 Ferric ion plays an important role in living organism . In mammal,ferric ion is carried by transferrin to different organs . Through the binding of transferrin to its receptor on the cell membrane ,ferric ion is entrapped into the cells. This study focused mainly two aspects . First, we prepared a horseradish peroxidase conjugated transferrin antiboby , which was used well to measure the distribution of transferrin among different organs including brain , lung ,heart, liver , pancreas ,spleen ,kidney ,testis,epidymis semina1 vesicle ,anterier prostate, ovary,uterus. Tansferrin was found in any one of the organ through the amount of transferrin in each organ varied . It was of interest to note that transferrin was very rich in both seminal vesicle and seminal fluid. Second , we attempted to purifyed antibody for transferrin receptor of mouse reticulocytes. The preliminary results where not succeed but encouraging. |
