摘要: | 利用實驗室已有的三種野生種菸草(Nicotiana plumbaginifolia, N. sylvestris, N. otophora)單倍體葉肉原生質體融合一起的產物為材料,分別以細胞學(染色體組成),生物化學(同功異構)和形態學的方法,鑑定是否有體細胞雜種的產生。結果發現在所挑取30塊癒合組織分化出之64棵植株中,全部都是雜種。其中包括三種親本染色體組同時出現在一細胞者。由於原生質體融合之後並未加任何人工篩選,而所得全為雜種,因此我們相信種間的雜種可能有雜種優勢的存在。 在這些雜種中挑取不同親本組合所作的電泳分析,計有5種酵素的同功異構?可以鑑定體細胞雜種,分別為peroxidase, malate dehydrogenase, superoxide dismutase, aspartate aminotransferase和aryl esterase。所得雜種並移植於溫室作形態上的比較,雜種的花、葉等特徵大部分介於親本之間。 By the use of polyethylene glycol (PEG), the mesophyll protoplasts from haploid plant of three Nicotiana species (Nicotiana plumbaginifolia, N. sylvestris, N. otophora) were induced to fuse (already done by studio patern). All the 64 plants regenerated from 30 calli of the fused result, have been identified as somatic hybrids by chromosome identification of root tip cells. Among these, some hybrids contain three different genomes. Because no selection was performed for hybrid cells and all the regenerated plants were hybrids, it is believed that the somatic hybrids exhibited heterosis and readily produced shoots. We have selected 4 different hybrid combinations for polyacrylamide gel electrophoresis (PAGE). Of the 17 enzymes we tried, only 5 enzymes (Peroxidase, Malate dehydrogenase, Superoxide dismutase, Aspartate aminotransferase and Aryl esterase) could be used as biochemical markers for somatic hybrid identification. After transfer of rooted shoots to soil flowering plants were obtained in all combinations. The morphology of most of the somatic hybrids are intermediate between the respective parental plants. |
