摘要: | 以水稻、台南五號(Oryza Sativa L. var. Tainan 5)懸浮培養細胞?材料來源.分離出水稻原生質骨體。得到1×10^7個、克鮮重的的高比例。以1.2% agarose 凝固的KPR培養基斷片培養(agarose segment), 30天后。 原生質體分裂成小細胞團的比例(plating officicncy)?0.2%。 分離出之原生質體經45℃ 8分鍾。碎冰10分鍾處理。會使原生質體活性百分比提高.而此原生質體在培養30天。Plating efficiency 提高了一倍;而40℃ 2個小時的熱休克處理同樣都會使水穩細胞(intact coll)生長速率提高。 以SDS Polyacrylamidc 電泳分析比較45℃與40℃熱休克,處理後蛋白質合成的變化。45℃組所沒有的。而在70KD、37KD、處蛋白質的量比40℃組多。而40℃組.則是-31KD蛋白質是45℃組所沒有的.進一步以變向電泳分析。看出在57KD處有5種蛋白質。34KD處有一種蛋白質是40℃組所沒有的.而在70KD處有4種蛋白質的合成量比40℃組多.推測這57KD、34KD、70KD三群蛋白質在原生質體的培養與分裂能力的提高上扮演重要的角色。 Protoplast isolated from rice (Oryza sativa L. Tainan 5) com suspension culture get the high ratio or plating frequency as 0.2%. while cultured with 1.2% agarose solified KPR meatiom after 30 days. Heat shock (45℃ for 8 min) treated proto plasts exhibited an effective doubling in both the frequency of plated protoplasts dividing after 14 days and in plating efficiency compared with untreated protoplasts, white the heat shock treatment (40℃ for 2hr) didn't have the same results. However. for rice intact cells, both of the heat shock treatment did increase the growth rate. The protein pattern of cultured rice proto plasts treated with 45℃ (8min) was compared to that treated with 40℃ (2hr) on one dimentional SDS-polyacrylamide gels. Two protein bands 37.000 and 70.000 dalloms were much more abun dant in the 45℃ treated protoplasts, and four protein dands (57.000、51.000 34.000 and 8.000daltons) were unique to the 40℃ treated proto plasts. Using two-dimentional gel analysis. there were four hsps (40.000 daltons) were much more abundant in 45℃ treated protoplasts, and five hsps (34.000 daltons)、 one hsp (57.000 daltons) were unique to the 40℃ treated proto plasts. We suppose that these three group proleins (34.000, 57.000, 70.000 daltons) may play an important role in the enhancement of protoplast division in rice. |
