具有致死性的蛇毒蛋白,Mojave toxin, 是由美洲響尾蛇(Mojave rattlesnake, Crotaluo Acutulatuo cutulatuo)的毒液純化得到,再經由陽離子交換樹脂,Sp-Sephadex C-25,可將其兩個次單元分離。具有毒性與磷脂?活性的酸性次單元,稱?Mojave toxin A,經由還原烷化將雙硫鍵打斷,以析柱Sephadex G-50,得到兩個部份。整個酸性單元及經由Sephoder G-50的兩個部分,本實驗D ABITC/PITC 雙偶合的方法,決定其N-端胺酸排列順序與比較。
具有毒性與磷脂?活性的鹼性次單元,稱?Mojave toxin B,與還原烷化後的鹼性次單元R(M-Mojave-toxin B)。同作N-端胺基酸排列順序為:His-Leu-Leu-Gln-Phe-Asn-Lys-Met-Ile-Lys-Phe。Mojave toxin B以BRCN切判後,以Sephadex G-25分離得到兩個部分為:CNBI 及CNBII/III,而其中CNBII/III含雙硫鍵,還原烷化後,同樣以Sephadex G-25得CNBII及CNBIII兩個部分;而它們的部分胺酸排列順序被決定出來。
