dc.description.abstract本實驗目的在探討鯉魚頭腎蛋白激?A (cAMP-dependent Protein Kinase,簡稱pkA)的特性,於實驗過程中,也發現頭腎組織含有可抑制pkA活性的抑制物,即蛋白激?A抑制蛋白(cAMP-dependent protein kinase inhibitor ,簡稱pkI),因此本實驗也對pkI進行初步探討。
頭腎於4mM EDTA-15mMβ-mercaptoethanol (pH7.0)以均質機勻碎,離心棄沈澱得粗分均質液,再以DE-52滌流之。發現pkI活性見於120-200 mM及360-450 mM NaCl滌流區。本實驗曾兩次層析頭腎組織的pkA活性,發現兩次結果頗有差異:第一次組織所得之pkA活性全不受cAMP的影響,而第二次的層析,則見有仰賴cAMP的pkA,此項結果顯示鯉魚頭腎組識中含有兩種型式的pkA。
鯉魚頭胃組織的pkI也可用DE-52管柱層析之,pkI活性見於150?300mM CH3COONa滌流區,所得pkI不具去磷酸?之活性。
