dc.description.abstract | 本研究的目的擬建立感染性胰臟壞死病毒持久性感染吳郭魚卵巢細胞株(IPNV Persistent infectious TO-2 cell line)並研究其生長特性。 利用自鰻腺分離出來的感染性胰臟壞死病毒(FV-1229)感染吳郭魚卵巢細胞株(TO-2),可以?立病毒持久性感染細胞株,命名為P1/TO-2。其特徵為可持久性的釋放病毒顆粒(P1病毒)於培養基中,對同源病毒具有抗性,但可接受異源病毒的攻擊性感染,由電顯觀察及螢光免疫實驗發現P1/TO-2細胞株中只有少數細胞含有病毒抗原。 由外部形態及對胞內LDH, MDH, G6PDH和Esterase四種同功?的電泳分析,均無法區分P1/TO-2及TO-2細胞株。電子顯微鏡觀察雖發現P1/TO-2細胞株中只有少數細胞的細胞質含有病毒顆粒,但在細胞碎片中則常可發現病毒群,此結果顯示P1/TO-2細胞株培養基內的病毒是由少數含病毒顆粒的細胞破裂而來。 P1病毒和原來的FV-1229病毒均可為抗FV-1229兔血清完全中和,顯示二者在血清型上並無差別,其他如電顯觀察,溫度適應性質實驗及病毒寄主範圍實驗,亦無法判斷二者的不同。然二者在增殖過程中均有自我幹擾現象發生,實驗推論病毒族群中含有缺陷性幹擾顆粒(Defective interfering particle),此種顆粒可能為P1/TO-2細胞株建立及維持生長的主要因數。用含有抗FV-1299兔血清的培養基培養P1/TO-2細胞株,無法治療甚至降低細胞株中含病毒的細胞數目。 | zh_TW |
