摘要: | 本實驗的目的是探討吳郭魚滲透壓調節與緊迫反應之 HSP90 的相互關係。吳郭魚加熱10℃與轉移至20ppt 海水環境,可能改變鰓上 HSP90 與 Na+,K+-ATPase 蛋白質表現量和血液滲透壓與 Na 離子濃度。另外,在加熱與海水轉移的過程中利用 HSP90 專一性抑制劑 geldanamycin 以證實 HSP90 與吳郭魚滲透壓調節的關係。實驗結果發現,加熱處理組之魚體體內HSP90 與 Na+,K+- ATPase 蛋白質表現量高於對照組,而血液滲透壓與 Na 離子含量低於對照組。加熱處理組對後續海水轉移有較佳的適應能力,包括刺激Na+,K+-ATPase 。表現與穩定血液滲透壓。吳郭魚同樣在加熱及轉移海水後,以HSP90 專一性抑制劑 geldanamycin 處理魚體,體內 Na 離子總量低於對照組。這些結果顯示,加熱所產生的 HSP90 蛋白質,可能與魚類海水轉移的滲透壓調節機制有關。 The purpose of the present study is to explore the relation between osmoregulation and HSP90 in tilapia. Effects of +10°C heat shock and transfer to 20 ppt SW on the expression of gill HSP9O and Na+, K+-ATPase as well as plasma osmolarilty and body Na+ content in tilapia (Oreochromis mossambicus) were examined. Moreover, specific inhibitor of HSP90, geldanamycin, was used to reconfirm the relationship between HSP90 and osmoregulation in tilapia during heat shock and SW acclimation. The protein expression of HSP90 and Na+, K+-ATPase in heat shock group was higher than that in control group, and the value of osmolarity and Na+ concentration in heat shock group was lower than that in control group. The rapid heat shock treatment is helpful for heat shock group stimulating Na+, K+-ATPase and consequently, enhancing salinity acclimation. Immersion of the specific inhibitor of HSP90, geldanamycin, caused tilapia to show a significantly higher Na content than the control after heat shock and SW transfer. Based on these data, we concluded that HSP90 was influential to the osmoregulation mechanisms in fish during SW acclimation. |
