dc.description.abstract數百萬年來,脊椎動物生活在各種入侵者的環境之中;這些入侵者包含了病毒,細菌及各種寄生蟲。免疫系統演化的首要目的是為了對抗外來的致命入侵者。免疫反應在對抗外來抗原之基本特徵為;針對特定抗原有其專一性。其中,第一類、第二類 MHC 及 CDI 分子是包含在 T 細胞反應中的三種主要抗原呈現分子,同時他們和細胞激素的釋放有關聯。
肺炎雙球菌是一種革蘭氏陽性細菌。肺炎雙球菌的細胞壁主要是由?聚醣所組成:其中的聚醣鏈是由 N-乙醯葡萄糖胺及 N-乙醯胞壁酸的殘基交錯所組成,糖鏈間的交互聯結是藉由勝?鍵及乙醱胞壁酸結合完成。細胞壁的多醣是一種包含了磷脂膽鹼殘基的台口酸,而且也是藉由磷酸二脂鍵結到 N-乙醯胞壁酸和?聚糖相連結。另一種重要的結構是脂台口酸(或稱為福氏抗原『Forssman antigen』)是一種台口酸相似於細胞壁中的多醣且具有額外的共價鍵連結到脂質的部份。
在一篇研究中,我使用了化學性質已明的肺炎雙球菌細胞壁成份去刺激取自腹腔的巨噬細胞。我發現了下列的現象: l )當用萃取出來的細胞壁刺激時,第一類 MHC 分子 mRNA 的表現會提升; 2 )當台口酸刺激巨噬細胞時,CDI 的 mRNA 表現增加, ; 3 )當受到萃取出來的細胞壁刺激時,間白素-Iα mRNA 的增加,是; 4 )用萃取出來的細胞壁及?聚糖刺激時,則會促進間白素-6 的 mRNA 的表現; 5 )間白素-12 mRNA
的表現則會受到肺炎雙球菌細胞壁所有成份的影響; 6 )相對地,間白素-l β , 2 , 4 及幹擾素-γ的 mRNA 則不會受到細胞壁成份的刺激而發生重大改變。

dc.description.abstractVertebrates, over the course of millions of years, have been continuously exposed to invasion by viruses, bacteria, and other parasites. The immune system has evolved primarily to deal with these potentially lethal invaders. The most basic feature of the immune response to a foreign antigen is its specificity for that particular antigen. Three antigen-presenting molecules, MHC class I, MHC class II, and CD1, are implicated in T cell responses and together they are associated with the release of cytokines.
Streptococcus pneumoniae is one type of gram-positive bacteria. Pneumococcal cell wall is composed mainly of peptidoglycan: glycan chains of alternating N-acetylglucosamine and N-acetylmuramic acid residues, cross-linked to each other through peptide side-chains via N-acetylmuramic acid. The cell wall polysaccharide, a complex teichoic acid containing phosphocholine residues, is also attached to peptidoglycan via phosphodieaster bond to Nacetylmuramic acid. Another important structure is lipoteichoic acid or Forssman antigen, a polysaccharide similar to teichoic acid with additional covalently attached lipid moiety.
In this study, I used chemically well-defined components from pneumococcal cell wall to stimulate the peritoneal macrophages. I found that: 1) The expression of the MHC class I mRNA was elevated by the cell wall materials. 2) CD1 mRNA expression was stimulated by lipoteichoic acid. 3) The IL-lα mRNA increased when macrophages were stimulated by cell wall materials. 4) The expression of IL-6 mRNA was elevated by using cell wall materials and peptidoglycan. 5) The expression of IL- 12 mRNA is influenced by all components from streptococcal cell wall. and 6) The mRNA expressions of IL-1β, 1L2, 1L4, and IFN- γ did not change significantly by all of the cell wall components.
