dc.description.abstractEscherchlia coli J 5-3可以與變種植物病原菌Pseudomonas solancearum 68st; Xanthomonas oryzae 604; X. eampestris 11及X. campestris pv. citri 47進行conjugation.□□可轉入質體RP,進入這些植物病原菌,其轉入□□□(transfer frequeney)依細菌之種類,□有不同的結果,P. Solancearna 6881為1.781×10^(-6); X. oryzae604為7.19×10^(-6);X. eampestris 11為1.04×10^(-2); X. campestris pv. Citri 17為2.09×10^(-3),同時在conjugation□□□□□,□□度□□□。用ethidium bromide及acridine orange □□□□P.soiancearum 68st transconjugant之質體RP; 其□□試驗□度與處理時間,呈現正比的關係。□□□方式確定質全能轉入與去除外,抽取P.soiancearum 68st transconjugan之質體,作agarose電冰分離,可以和E.coli J5-3之質體RP,有相同的電冰□,□□□去除其□處理之transconjugant,則此電冰帶不存在,如此更確定質體之轉入與去除之正確性。
比較P. solancearum 68st之wild type; transconjugant:及時去除□□處理之transconjugan的病原性,接種之植物增色能生長□□,對此P. solancearum 68st之病原幾乎是不存在的質體上,但是transconjugan所產生的病體經較轉□,□□細菌生長狀態transconjugant無法達到wild type之能□□□。X.oryzae 604 transconjugant □能生起□□產生物質,但用電之的問題後移,研究細菌生長狀態,乃有緩慢之情形,此□X.campestris 11及X.campestris pv. citri 47在細菌生長之研究時,也發現他們的trans-conjugant有類似的情形,因此質體PR4之轉入,可能影響細菌之生長狀態。

dc.description.abstractTranster of plasmid RP4 in some phytopathogenic Bacteria and Its Relation to Their Pathogenicities
The plasmid RP4 can transfer from Escherichia coli 85-3 into several phytopathogenic bacteria: Pseudomonas solaaacearum 68st, Xanthomonas oryzae 604. X. campestris 11 and X. campestris pv citri 47. the frequencies of temasfor for these bacteria are 1.78×10^(-6), 7.19×10^(-6), 1.48×10^(-2) and 2.09×10^(-3) respectively. The frequency of transter depends on baeterial species. Conjugation conditions. Using ethidium bromide and acridine orange. The plasmid RP4 in the transconjugant of P. solanacerum 68st can be cured. The plasmid transferred into P. solanacearum 68st and eured from the transconjugant from the sample of P. solanacearum 68st transconjugant was detected .The posision of plasmid band was identical identical with the plasmid band obtained from the sample of E. coli J5-3.
The pathogenicities of the wild type, transconjugant and cured transconjugare from P. solancearum 68st and X. oryzae transconjugart form p. solanacearum 68st and x. oryzae 604 were comparod. Either transconjucant on oured transconjugant showed a typical symptom on inoculated plants. It appears that the pathogenicity is not existed in the plasmid. However, when the aransconjugant was inoculated, the delay of symptom expression was ebserved. When the growth of wild aype and transconjugant were compared. the growth of transconjugant was slower than than that of wild type. This result may explain the cause of the delay of symptom expression.
