dc.description.abstract | 本研究以半穴居的短尾鼩為對象,進行微隨體基因座選殖的工作,並利用多型性的微隨體基因座探討短尾鼩的親族關係。利用(AC)10、(TC)10、(CAC)5CA、CT(ATCT)6、(TGTA)6TG和CT(CCT)5六種探針,針對8400個菌株進行雜合反應,一共篩選出46個重複次數大於5次以上的微隨體基因座,其中有21個以AC為主、19個以TC為主、2個以CCT為主和額外篩選出4個以其他重複為主的微隨體基因座。進一步以微隨體基因座兩側的序列進行PCR引子的設計,發現其中11個基因座在短尾鼩族群中呈現高度的多型性,有些基因座的對偶基因甚至高達20個,且Ho和He的範圍分別介於0.33-0.92和0.82-0.96之間。利用這些多型性的微隨體基因座探討36隻短尾鼩的親族關係,並依分佈範圍的縮小將之區分成3個不同的族群,發現三個族群觀察的親族關係和模擬哈溫平衡下、逢機交配的族群並沒有差異,表示族群內可能無近親交配的情形,且使得族群的個體間並沒有明顯的親族關係,但代表親族關係的R值,隨著採集地點範圍縮小仍有增加的趨勢。 | zh_TW |
