摘要: | 牛樟芝(Antrodia cinnamomea)為臺灣特有之藥用真菌,存在多種活性物質,其中已證實多醣具有免疫調節活性及護肝活性,然其結構特徵尚不清晰。本研究分析牛樟芝菌絲發酵物凍乾粉末中含有15.5%的葡萄聚醣,包含2.6% (1,3)-β-D-葡萄聚醣、10.9% (1,4)-β-D-葡萄聚醣、2.6% (1,6)-β-D-葡萄聚醣與1.1% 可以消化的α-D-葡萄聚醣。已知菇蕈類多醣體中以具有β-(1,6) 側支鍵結葡萄糖的(1,3)-β-D-葡萄聚醣為其生物活性指標,本研究利用膠體過濾層析得到牛樟芝多醣及(1,3)-β-D-葡萄聚醣之分子量分布。牛樟芝菌絲發酵物中水溶性膳食纖維以非消化水溶性多醣為主,其主要糖組成為半乳糖(58.67%)、葡萄糖(28.41%)、甘露糖(7.91%)與岩藻糖(2.79%),以及少量的阿拉伯糖與木糖。利用DEAE陰離子交換層析管柱劃分非消化水溶性多醣為三個區分,三個區分皆含有岩藻甘露半乳聚醣、(1,6)-β-D-葡萄聚醣及(1,3; 1,6)-β-D-葡萄聚醣;鍵結分析中半乳糖基以末端與1,6 為主要鍵結並有2,6分支點;甘露糖以末端及1,2為主要鍵結;岩藻糖以末端形式存在為主,推測其為甘露糖以1,2鍵結穿插在1,6鍵結的半乳糖主幹中,分支處接有岩藻糖或甘露糖的岩藻甘露半乳聚醣;葡萄糖基以末端、1,3與1,6 為主要鍵結並有3,6鍵結之分支點,推測其來自(1,6)-β-D-葡萄聚醣及(1,3; 1,6)-β-D-葡萄聚醣。將酸性醣之羧酸基還原後,發現區分F3為帶有葡萄糖醛酸之醣蛋白,葡萄糖醛酸鍵結形式以末端、1,3及3,6分支點為主。本研究使用四級銨鹽選擇性沉降法濃縮牛樟芝菌絲發酵物水溶性多醣中酸性糖的比例後再以陰離子交換層析管柱進行後續劃分,可改善純化分離流程中陰離子交換層析耗時及承載量限制的缺點。 Antrodia cinnamomea has been considered as a potential medicinal mushroom in Taiwan. It is immunomodulatory attribute has been associated with its polysaccharides. This study analyzed the polysaccharide profile of whole mycelium fermentation mass of Antrodia cinnamomea including mycelia and broth. The selected sample contained 15.5% glucan, which consists of two well-known bioactive glucans, (1,3)-β-D-glucans (2.6%, on dry-basis) and (1,6)-β-D-glucans (2.6%), as well as (1,4)-β-D-glucans (10.9%) and digestible α-D-glucans (1.1%). The water-soluble dietary fiber of the sample composed of indigestible water-soluble polysaccharides (IDPS). The sugar composition of IDPS was found to be galactose (58.67%), glucose (28.41%), mannose (7.91%), fucose (2.79%) and a small amount of arabinose and xylose. The identified components of IDPS included β-(1,3; 1,6)-D-glucans (0.05% on polysaccharide-basis) and β-(1,6)-D-glucans, fucomannogalactans, a peptidoglycan and a glycoprotein through enzymatic assays and DEAE-anion exchange chromatography. The the linkage patterns of fucomannogalactans included terminal-galactosyl, 1,2-galactosyl, 1,3-galactosyl, 1,4-galactosyl, 1,6-galactosyl, 2,6-galactosyl, 3,6-galactosyl, 4,6-galactosyl, 3,4,6-galactosyl, 1,2-mannosyl, terminal-mannosyl and terminal-6-deoxy-L-galactopyranosyl (Fuc). IDPS could be separated by DEAE chromatography into three fractions, F1, F2 and F3. The linkage analysis showed that the neutral fraction F1 is mainly fucomannogalactan and a small amount of (1,3; 1,6)-β-D-glucan; F2 and F3 is mainly (1,3; 1,6)-β-D-glucan and a small amount of fucomannogalactan. After the reduction of the carboxylic acid group of the acidic sugar, it was found that glucuronic acid was present in the F3, and the linkage form is terminal-glucuronic acid, 1,3-glucuronic acid and 3,6 glucuronic acid. A preparative procedure for glycoproteins of A. cinnamomea had proposed by using quaternary ammonium salt selective precipitation method. This study provides the polysaccharide profiles of fermentation mycelium mass of A. cinnamomea and the profiles can be used as a quality control tool for the development of polysaccharide-based functional food products in the future. |
