摘要: | MSI1-like (MSIL)蛋白是哺乳類動物中RbAp48蛋白的同源蛋白,這類蛋白通常沒有任何催化活性,但對於生物中生長發育卻扮演著重要的角色。在後生動物以及植物中,MSIL蛋白通常會和其他蛋白形成複合體並調節染色質的動態變化。在阿拉伯芥中,有五類MSIL蛋白,其中第四類蛋白稱做MSI4/FVE。MSI4/FVE會經由抑制開花負調控因子FLC影響植物開花。組蛋白去乙醯化酶HDA6是屬於RPD3類型的組蛋白去乙醯化酵素。在這篇研究中,我們證實了FVE可以跟HDA6相互作用,並且經由影響組蛋白去乙醯化(H3K9K14Ac)以及組蛋白甲基化(H3K4me3)抑制FLC的表現。我們也進一步研究了FVE是否也參與非生物性逆境反應。經由發芽率以及根長的實驗,我們證實了FVE也參與在ABA以及滲透壓的逆境反應,並且ABA反應相關的基因也在受到FVE的調控。綜合以上結果顯示出FVE/MSI4對於植物開花以及非生物性逆境反應都扮演著重要的角色。 MSI1-like (MSIL) proteins are homologous to the mammalian RbAp48 protein, which shows no catalytic activity but has specific function during development. In both metazoans and plants, MSI1-like proteins are part of several protein complexes involved in diverse chromatin functions. A plant homolog of RbAp48, MSI4/FVE, is involved in the autonomous pathway regulating flowering time by repressing FLC. HDA6 is a RPD3-type histone deacetylase. In this study, we show that HDA6 interacts with FVE and regulates flowering time by repressing FLC through decreasing H3K4 trimethylation and H3 acetylation. In addition, FVE was also involved in abiotic stress responses. Compared with wild-type, the hda6, fve-4 and hda6fve double mutant plants were hypersensitive to ABA and mannitol in seed germination and root length assays. Furthermore, the expression of ABA-response gene was changed in these mutants. Taken together, these data suggest that FVE/MSI4 plays an important role in flowering and abiotic stress responses. |
