摘要: | 紅麴酒萃物 (red mold rice ethanol extracts, RMRE) 能抑制 squamous cell carcinoma-4 (SCC-4) 鱗狀上皮細胞癌細胞株生長,其百分之五十抑制濃度 (50% inhibition concertraction, IC50) 為116.0 μg/mL,而主要抑制癌細胞生長的成分為 monacolin K,然紅麴酒萃物中其他成分亦具有協同抑制癌細胞生長。以脂多醣 (lipopolysaccharide, LPS) 誘導RAW264.7 氧化發炎細胞模式實驗結果得知,紅麴酒萃物具有抑制氧化發炎效果,但並非由單獨由monacolin K 造成的,而是可能由 monacolin K、citrinin、monacin、ankaflavin 等其他物質導致。因此利用二甲基苯蔥 (7, 12-dimethyl-1,2- benz[a]anthracene, DMBA) 誘導倉鼠口腔癌化動物模式,塗抹紅麴酒萃物,探討評估紅麴對於口腔癌病變的影響。實驗結果顯示,DMBA會造成體內活性氧化 (reactive oxygen species, ROS)、一氧化氮 (nitric oxide, NO)、前列腺素 E2 (prostaglandin E2, PGE2) 等氧化、發炎因子上升,藉由塗抹紅麴酒萃物處理後,可抑制 DMBA 誘導的氧化、發炎現象,因此紅麴酒萃物可能藉由抑制發炎、氧化作用來降低口腔癌的癌化。除此之外, DMBA 誘導癌化過程中,Cu、Zn、Ca、Mg、Cu/Zn 數值比正常組別偏高,且在第十週時口腔癌由原位癌惡化至侵襲癌,其毛髮中之微量元素,亦有急劇之變異。雖微量元素之測定仍尚未如生物活性指標來得精確,但其生理之意義仍有進步探討研究之必要。 The ethanol extract of red mold rice (RMRE) can inhibit the growth of squamous cell carcinoma-4 (SCC-4) cells (IC50 = 116 μg/mL), and monacolin K is the main component in RMRE to exert the inhibition of cancer cell growth. Moreover, RMRE inhibited lipopolysaccharide-induced oxidative stress and inflammation in RAW264.7 cells. And the anti-oxidation and anti-inflammatory effects were not only due to monacolin K, but supposed to be a synergic effect among monacolin K, citrinin, monacin, ankaflavinn, and others. Furthermore, DMBA induced oral cancer in hamster model was also established to estimate the anti-cancer effect of RMRE. RMRE inhibited oxidative and inflammatory stress to reduce the carcinogesis of oral cancer through down-regulation of the increased levels of reactive oxygen species, nitro oxide, and prostaglandin E2 induced by DMBA. In addition, we found the Cu, Zn, Ca, Mg and Cu/Zn values were significantly elevated in hairs of DMBA induced oral cancer hamsters. Further studies are necessary to investigate the relationship between elevated heavy metal trace elements and the progression of carcinogenesis. |
