dc.description.abstract | 台灣穿山甲(the Formosan Pangolin, Manis pentadactyla pentadactyla),是鱗甲目(Pholidota)中位於亞洲的四個種(four Asian pangolin species)裡面中國穿山甲(the Chinese Pangolin, Manis pentadactyla)的特有亞種,鱗甲目在哺乳動物的系統分類上,究竟是跟食肉目(Carnivora)比較接近,還是跟貧齒目(Xenarthra)比較接近還有一些爭議。
本博士研究由台灣穿山甲出發,先決定它的核型(Karyotype),定出染色體示意圖(Ideogram),然後選殖它的Sry(雄性性別決定基因)基因,證明台灣穿山甲在核型上跟分佈在中國大陸的華南穿山甲(Manis pentadactyla aurita)在異染色質(heterochromatin)分佈以及核仁組成區(Nucleolus Organizing Region, the NORs)的數目上略有不同,兩者確實可以看做中國穿山甲的兩個特有亞種。為了解答鱗甲目的系統分類問題,我同時也選殖貧齒目中的二趾樹懶(the two toed sloth, Chloepus didactylus)的Sry-HMG (High Mobility Group domain of Sry),得到它的核苷酸序列,與已知的物種作親緣關係分析,得到鱗甲目跟食肉目比跟貧齒目要來得接近的結果。我也同時利用細胞遺傳學的方法首先證明所選殖到的台灣穿山甲Sry基因確實位在Y染色體上,接著使用競爭性全染色體塗抹(competitive chromosome painting)的策略,成功發展出一種方法標示出台灣穿山甲的性染色體X和Y;接著利用改良過的全基因組原位雜合反應(Genomic in situ Hybridization, the “GISH”),證明台灣穿山甲的基因組相似度跟家犬比跟二趾樹懶接近;最後,我參考原先用於人類腫瘤遺傳研究的比較性基因組雜合反應(Comparative Genomic Hybridization, the “CGH”),進一步將之修正為將二趾樹懶的DNA和家犬的DNA分別標定上紅色螢光分子以及綠色螢光分子(或相反,即dye swapping),將之競爭性結合回公的台灣穿山甲的細胞分裂中期的細胞展片(metaphase spreads)上,我得到強有力的證據證明台灣穿山甲的基因組相似度跟家犬比較相近,跟二趾樹懶比較遙遠。 本博士研究解決了鱗甲目在哺乳動物系統分類地位爭議,並且選殖或放大到以往沒有報告過的兩個序列(台灣穿山甲的Sry基因全長以及二趾樹懶的Sry-HMG核苷酸序列),更重要地,本研究開發出兩種新的方法,可以比較哺乳動物兩個物種之間、或者三個物種之間全基因組的相似度,替細胞遺傳學在演化生物學上的應用,開啟了全新的一個方向。 | zh_TW |
