dc.description.abstract | 澱粉是植物體能量分子的主要儲藏型式,在眾多澱粉降解酵素中,β-澱粉酶屬於澱粉外切酶,廣泛存在於作物種子與植株各部位細胞之細胞質、液胞與葉綠體中,表現強度會受到植物生長發育時期、非生物性逆境、糖類、與植物荷爾蒙等多種因子所調控。早期研究大多針對小麥族植物之種子發育與萌芽時期的β-澱粉酶活性角色,近年來利用阿拉伯芥突變種為材,則發現葉綠體型的β-澱粉酶在暫存性澱粉降解作用中佔有極重要的地位,並且可能參與了植物對抗低溫逆境的機制。隨著水稻基因組定序計畫的完成,解碼的水稻栽培種日本晴提供了研究單子葉(非小麥族)植物β-澱粉酶系統絕佳的研究材料。搜尋已公佈的水稻日本晴基因組資料庫,發現有十個可能的β-澱粉酶基因,本文將之編號為OsBMY1~10。以其胺基酸序列與已知之玉米、阿拉伯芥等的β-澱粉酶相比對,可將這些β-澱粉酶大致分為兩大群,其中一群與葉綠體型的β-澱粉酶較為相似。利用RT-PCR分析發現OsBMY6在萌芽中胚乳並無表現,如同早期文獻所記載;而序列未包含完全保守性區域的三個基因,OsBMY2、OsBMY3和OsBMY9也偵測不到基因表現。OsBMY1在各個測試部位都有明顯表現,其餘基因表現則有組織差異性:OsBMY4主要在花部表現,OsBMY5、8主要在地上部之葉片表現,而OsBMY7、10則在癒傷組織與生殖器官表現。針對大量表現的OsBMY1基因研究發現,在癒傷組織與小苗地上部都有OsBMY1蛋白質表現,又以地上部為多,且可對應回Native-PAGE澱粉酶同功酶的活性分析圖譜中的S1活性條帶。同時,藉由分離葉綠體內蛋白質進行免疫轉印也證實OsBMY1為葉綠體型的β-澱粉酶之一。此外,OsBMY1在地上部的表現量還會受到高溫、鹽逆境以及ABA處理而增加。在利用農桿菌轉入OsBMY1基因反義股的水稻癒傷組織細胞中,OsBMY1的mRNA表現量被抑制為野生型的一半,β-澱粉酶活性也下降,對於熱與鹽逆境的抗性較低,意味著OsBMY1可能參與水稻對抗逆境的機制。 | zh_TW |
