摘要: | 本研究之目的為探討11種樟科(Lauraceae)植物葉部抽出物之生物活性,評估抗真菌活性、抑制黃嘌呤氧化酶活性、抗氧化活性及抑制蕈類酪胺酸酶效果,並以斑馬魚為模式生物進行抑制黑色素生成活性的評估,期盼提升樟科植物天然物的用途。結果顯示11種樟科植物葉部抽出物不具有抑制黃嘌呤氧化酶活性,對於抗真菌的效果亦為有限。抗氧化活性分析結果發現,變葉新木薑子(Neolitsea aciculate)、小芽新木薑子(Neolitsea parvigemma)、鹿皮斑木薑子(Litsea coreana)與長葉木薑子(Litsea acuminata)葉部抽出物含有較多之總酚類化合物與總黃酮類化合物,在還原力、自由基清除活性與總抗氧化能力等評估皆具良好效果。
在蕈類酪胺酸酶評估中,以小芽新木薑子、鹿皮斑木薑子、內苳子(Lindera akoensis)及瓊楠(Beilschmiedia erythrophloia)等抽出物顯示最佳抑制活性,經酵素動力學分析,4種抽出物均屬競爭型抑制物。而在抑制斑馬魚黑色素生成效果的評估中,效果較佳的抽出物依序為長葉木薑子 (IC50 = 71.36 μg/mL) > 內苳子 (IC50 = 114.46 μg/mL) > 香桂(Cinnamomum subavenium) (IC50 = 117.32 μg/mL) > 五掌楠 (Neolitsea konishii) (IC50 = 122.19 μg/mL) > 樟樹(Cinnamomum camphora) (IC50 = 127.12 μg/mL) > 瓊楠 (IC50 = 188.16 μg/mL),顯示長葉木薑子抽出物對於減少斑馬魚黑色素生成的表現最佳。綜合上述結果得知11種樟科植物中,變葉新木薑子、小芽新木薑子、鹿皮斑木薑子與長葉木薑子抽出物的抗氧化效果最佳,長葉木薑子抽出物的美白效果最好,未來可針對其活性成分加以探討,期盼能開發為良好之抗氧化劑及美白產品,增加林木天然物的利用價值。 The purpose of this study was to evaluate the bioactivities, including antifungal activity, xanthine oxidase inhibitory activity, antioxidant activity, antityrosinase activity, and melanogenesis inhibition activity in zebrafish (Danio rerio), of leaf extracts of 11 selected Lauraceae plants. Based on the results, 11 Lauraceae leaf extracts didn't possess the effect on antifungal and xanthine oxidase inhibitory activities. Leaf extracts of Neolitsea aciculate, Neolitsea parvigemma, Litsea coreana and Litsea acuminate contained the higher amount of total phenolic contents and total flavonoid contents, and these extract had superior antioxidant activities on reducing power, free radical scavenging activity, and total antioxidant capacity. N. parvigemma extract, L. coreana extract, Lindera akoensis extract and B. erythrophloia extract exhibited the better mushroom tyrosinase inhibition activity. In antimelanogenesis assay, six leaf extracts possessed the higher melanogenesis inhibition activity in zebrafish with a decreasing order of L. acuminate > L. akoensis > Cinnamomum subavenium > Neolitsea konishii > C. camphora> Beilschmiedia erythrophloia, and L. acuminate leaf extract exhibited the highest melanin inhibitory activity in zebrafish. Accordingly, among 11 Lauraceae leaf extracts, N. aciculate extract, N. parvigemma extract, L. coreana extract, and L. acuminate extract shows the best antioxidant activity, and L. acuminate extract exhibited the best whitening effect, these extracts are worthy to do further investigation on their active constituents and with the potent to be antioxidant agents and skin whitening products. |
