dc.description.abstract | 鉈是新興汙染物,且是一種容易被作物所吸收利用、具有劇毒的重金屬,若水稻為鉈所污染,將對食品安全造成莫大威脅。由於目前關於水稻對鉈的吸收、運輸及耐受性之分子機制都仍不清楚,本研究希望能尋找到水稻中與鉈耐受性相關的數量性狀基因座。本研究使用對鉈耐受的臺農67號與對鉈敏感台中在來1號做為親本雜交產生的198個重組自交系 (RILs) 為材料,此RILs已經由RAD定序方法分析基因型,共有1773個SNP多型性分子標記。在對照組及鉈處理下測量植株株高、地上部鮮重、乾重及受傷指數,並以這四個性狀進行數量性狀基因座定位,並定logarithm of the odds (LOD) 門檻值為3.4。在對照組處理下僅找到1個QTL區域qSLC3 (shoot length under control),在鉈處理下於第1、2、3、6與7號染色體上一共找到7個數量性狀基因座,其中位在3號染色體上的qSLT3 (SLT,shoot length under Tl treatment ) 與qSLC3定位在相同的區域,具有最高的LOD值10.58及最高的phenotypic variation explained (PVE) 值12.84%,其他6個QTL依LOD值排序為qSLT7.2、qSDWT2 (SDWT,shoot dry weight under Tl treatment)、qSFWT7 (SFWT,shoot fresh weight under Tl treatment)、qSDWT6、qSLT7.1以及qSLT1。此外,在鉈含量測定中發現,對鉈耐受的水稻品系在地上部相較對鉈敏感的品系累積較少的鉈,在地下部則沒有差異,顯示水稻可能藉由累積較少的鉈在地上部以減輕鉈毒害。後續研究可選擇qSLT3區間內所包含的33個基因為首要候選基因。 | zh_TW |
dc.description.abstract | Thallium (Tl) is an emerging toxic contaminant from electronic industries in the world. Tl is one of heavy metal and it is easily to be absorbed by crops. It will pose a great threat to food safety once rice is contaminated by Tl. The molecular mechanism about Tl absorption, transportation, and resistance in plants is not clear. Therefore, we aimed to map QTLs related to Tl tolerance in rice. In this study, we conducted QTL mapping against shoot length, fresh weight, and dry weight under control and Tl treatment by using 198 recombinant inbred lines (RILs) derived from the cross of Tl - tolerant TNG67 and Tl - sensitive TN1 rice cultivars. The single nucleotide polymorphism of these RILs have been identified by restriction site associated DNA sequencing analysis, and 1773 SNP marker were found. The LOD threshold for QTL is 3.4. One QTL, qSLC3, was found under control treatment, and 7 QTLs were found under Tl treatment. Among 7 QTLs, qSLT3 on chromosome 3 showed the highest LOD value, 10.58 and the highest PVE value, 12.84, and it overlapped with qSLC3. The others were qSLT7.2, qSDWT2, qSFWT7, qSDWT6, qSLT7.1 and qSLT1 sort by LOD value. We also found that Tl – tolerant lines showed lower Tl concentration than Tl – sensitive lines in shoots; however, there is no difference between two in root. It indicates that rice may reduce Tl toxicity by accumulating less Tl in shoot. For future study, the 33 genes in the interval of qSLT3 are the most potential candidate genes for Tl tolerance. | en |
