dc.description.abstract | 環境因子會透過不同層面來調控植物的生長與發育,光是植物最不可缺少的環境因子,而向光性是指植物會藉由生長素的調控沿著光方向生長,以提高植物在低光照環境下最大光合作用能力的反應。生長素和茉莉酸都是植物生命週期中重要調節作用的荷爾蒙,且生長素和茉莉酸的訊息傳導途徑有著相似的機制,以及前人的研究中顯示生長素和茉莉酸之間有著互相調控的關係;但向光性反應中光訊號的傳遞及生長素和茉莉酸調控途徑仍然不明確。 Far-Red Insensitive 219 (FIN219/JAR1)已被報導會參與在遠紅光及茉莉酸的訊息傳遞調控機制中,且其突變體 fin219-2在側邊藍光中具有向光性反應缺失的外表型;為了瞭解阿拉伯芥中FIN219、 PHOT (PHOTOTROPIN)和 NRL (NPH3/RPT2-LIKE)和向光性下游的信息傳遞在有無 MeJA (Methyl Jasmonate)的處理條件下,其三者的調控關係。我們檢測在側邊藍光中 fin219-2、 pGR219、 phot1、 phot2,以及其雜交株的外表型、基因表現以及蛋白質表現量;另外也檢測 MeJA處理後和未處裡的差異。實驗結果顯示,不管 fin219-2還是 pGR219在側邊藍光中都具有向光性反應缺失的外表型,且 MeJA會抑制植株彎曲的角度; DR5:GFP/fin219-2雜交株具有向光性反應缺失的性狀,其照光面與背光面的生長素不具有極性分布; FIN219扮演著調控 PHOT1與 PHOT2基因表現量,且 PHOT1與 PHOT2可能會共同抑制 FIN219基因及蛋白質表現; 另外 PHOT1與 PHOT2可能未直接的參與在 PHY A所調控的遠紅光訊息傳遞中。綜合目前研究結果推測阿拉伯芥的 FIN219可能參與在由 PHOT1所調控的向光性訊息傳導途徑中,且茉莉酸的調控也會影響向光性訊息傳導途徑。 | zh_TW |
dc.description.abstract | Environmental factors regulating multiple aspects of plant growth and development. Phototropism is controlled by auxin redistribution. Auxin and jasmonates are plant hormones that play important roles during many phases of the plant life cycle. Auxin and jasmonates use similar mechanisms in signal transduction pathways and they have some crosstalks that have been reported. How plants integrate the external light signal and endogenous Auxin and JA pathways during phototropism remains unknown. FIN219 has been shown to have roles in far-red light and jasmonate signaling and its mutant fin219-2 has a defect under unidirectional blue light. In order to understand the regulatory relationship among FIN219, PHOT, and NRL in MeJA and phototropism signal transduction in Arabidopsis, we examine phenotypic responses, mRNA and protein levels in fin219-2, pGR219, phot1, phot2 and their crossed mutants treated with or without MeJA under unidirectional blue light. Both fin219-2 and pGR219 had defect in phototropism under unidirectional blue light, and MeJA inhibited phototropism. DR5:GFP/fin219-2 had a defect in unidirectional blue light, which didn’t have asymmetric auxins distribution. Not only does FIN219 regulate PHOT1and PHOT2 mRNA levels, but also PHOT1 and PHOT2 redundantly inhibited FIN219. PHOT1 and PHOT2 indirectly participated in phyA-mediated signaling pathway. Our results revealed that FIN219 participates in phot1-mediated signaling pathway in phototropism, and JA signaling may influence phototropic responses in Arabidopsis. | en |
